To resolve these limitations about the Mycoplasma propagation, this research intended to optimize the ways to culture Mycoplazama. Also, this research intended to develop consensus polymerase chain reaction (PCR) which can examine the hundreds of Mycoplasma at once, and intended to develop species-specific PCR primers with high specificity and sensitivity which is able to identify M. hyopneumoniae and M. hyorhinis selectively. And also this research intended to develop the optimized infection method for the animal inoculation experiment with field isolated Mycoplasma strains and performed to compare the virulence of the field isolated Mycoplasma strains through animal inoculation experiment. As the results, the optimized Mycoplasma culture system was developed. Using the optimized media formula, M. hyopneumoniae and M. hyorhinis could be propagate quickly and efficiently. Also, consensus PCR for Mycoplasma detection was developed. The developed consensus primer pairs, MycoF and MycoR were designed specifically to amplify the 16S ribosomal RNA gene (rRNA) of Mycoplasma species by the optimized PCR system. The developed consensus PCR system effectively amplified 215 bp of Mycoplasma genus-specific region of 16S rRNA. And also the species-specific PCR primers against M. hyopneumoniae and M. hyorhinis was developed. The species-specific PCR primers were revealed high specificity and sensitivity against each M. hyopneumoniae and M. hyorhinis. These results could provide an efficient strategy and method for the rapid and accurate Mycoplasma diagnosis.
