Objective of these studies were to investigate the effects of seminal bacteria on embryo production in vitro and the sperm function in boars. To determine the efficiency of embryo production, boar semen were prepared with different concatenations of bacteria and used for in vitro fertilization. Also sperm viability and abnormality were investigated from the contaminated semen. Immature oocytes were cultured on in vitro maturation medium, NCSU-23,containing 10% porcine follicular fluid and hormones at 39℃, 5% CO2 for 20∼ 22 hr and then oocytes were further cultured without hormones for another 22 hr. Semen samples were prepared with 5 different levels of bacteria; 0, 3,000, 5,000, 7,000 or 10,000 cfu/㎖ and were used for in vitro fertilization. For sperm viability and abnormality test, semen samples were stained with fast green FCF (2%) and Eosin B (0.8%). The proportion of embryonic development of morula stage was significantly greater (p<0.01) in control group (43.7±8.6%) compared to other groups (36.6±6.0% in 3,000 cfu/㎖; 30.3±8.0% in 5,000 cfu/㎖; 23.6±7.7% in 10,000 cfu/㎖), indicating the negative relationship between developmental rate of embryo and number of bacterial contaminations in semen. Developmental rate of blastocyst stage was similar in groups (16.0±5.3∼ 18.4±5.7% in 0∼3,000 cfu/㎖), but rates were significantly lower (p<0.01) in 5,000∼7,000 cfu/㎖ group compared to control group (5.9±5.2∼9.0±5.4 vs 18.4±5.7%). Bacterial contamination level of 10,000 cfu/㎖ group showed the lowest embryonic developmental rate (p<0.01) compared to all other groups. The bacterial contamination as much as 10,000 cfu/㎖ level resulted in significantly lower sperm viability (85.7±10.0%, p<0.01) compared to contaminated levels below 7,000 cfu/㎖ (92.4±3.6∼99.3±0.3%). To investigate the prevalence of bacterial contamination in boar semen, seasonal comparison of contamination and select the effective antimicrobials to control contaminated bacteria. Semen samples were collected from 20 AI centers four times. Most of the fresh boar semen samples (94.8%) were contaminated, only 27.1% extended samples were contaminated with bacteria. In summer season, the rate of contamination of fresh semen was highest (100%)than other seasons (92.2∼94.8%). Gram negative (GN) bacteria were more frequently isolated from the boar semen than Gram positive (GP) bacteria. Effective microbial for GN were enrofloxacin, ceftiofur and gentamicin. Those were apramycin, bactriacin and ceftifut for GP. Because selected antimicrobials were not effective for all the contaminants, it will be suggested that mixture of two or three antimicrobials to control effectively.
