사람의 피부는 자외선, 오염된 공기, 화학제품 등 환경에 끊임없이 노출된다. 특히 자외선은 피부에 다양한 형태로 영향을 주어 주름, 잔주름, 피부거침, 피부건조와 같은 현상을 발생시켜 피부노화를 유발시킨다. 본 연구에서는 전 세계적으로 향나무과의 일종인 다년생 수목인 노간주나무(Juniperus rigida Sieb.) 추출물의 항산화효과와 미백 및 주름개선 효과에 대하여 검증하였다. 노간주나무 추출물의 항산화 측정으로 폴리페놀 함량을 측정한 결과 열수 추출물에서 71.3 ㎎/g, 에탄올 추출물에서 116.0 ㎎/g을 함유하고 있음을 확인하였으며, 플라보노이드 함량은 열수 추출물에서 17.7 ㎎/g, 에탄올 추출물에서 76.4 ㎎/g으로 나타났다. 전자공여능을 측정한 결과 노간주나무 열수 및 에탄올 추출물 1,000 ㎍/㎖에서 44.0%, 86.8%의 효과를 나타내어, 대조군으로 사용한 BHT와 유사한 효과를 나타내었으며, ABTS radical cation decolorization을 측정한 결과 1,000 ㎍/㎖에서 열수 추출물은 91.4%, 에탄올 추출물은 91.1%의 소거능을 나타내어 같은 농도의 ascorbic acid와 유사한 활성을 나타내었다. FRAP을 측정한 결과 노간주나무 열수 및 에탄올 추출물에서 1.83mM, 1.77mM으로 열수 추출물에서 더 높은 함량을 나타내었다. SOD 유사활성능을 측정한 결과 노간주나무 열수 및 에탄올 추출물은 1,000 ㎍/ml에서 43.4%, 32.3%의 활성을 나타내었으며, xanthine oxidase 저해활성능 측정 결과 열수 및 에탄올 추출물 1,000 ㎍/㎖에서 52.7%, 72.3%의 저해효과를 나타내었다. 미백효과를 확인하기 위하여 tyrosinase 저해활성을 측정한 결과 열수 추출물에서 49.4%, 에탄올 추출물에서 80.0%의 효과를 나타내어 열수 추출물에 비해 에탄올 추출물의 효과가 높게 나타났으며, 노간주나무 에탄올 추출물이 B16F10 흑세포종에 대하여 멜라닌 색소 생성 억제 및 tyrosinase, MITF, TRP-1, TRP-2의 억제 효과를 가지고 있는 것을 확인하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 노간주나무 열수 추출물 1,000 ㎍/㎖에서 10% 이하의 낮은 저해활성을 나타낸 반면에 노간주나무 에탄올 추출물의 1,000 ㎍/㎖에서 68.5%의 저해활성을 나타내었으며, collagenase 저해활성을 측정한 결과 1,000 ㎍/㎖에서 열수 및 에탄올 추출물은 44.9%, 97.2%로 에탄올 추출물의 경우 ascorbic acid (99.6%)와 유사한 효과로 높은 저해활성을 나타내었다. 또한 섬유아세포를 이용한 collagen 생합성량을 측정한 결과 노간주나무 에탄올 추출물 50 ㎍/㎖에서 151.52%의 생합성 촉진 효과를 나타내어 노간주나무 에탄올 추출물의 주름개선 효과가 우수하다는 것을 확인할 수 있었다. UVA에 의해 발현이 증가되는 MMP-1에 노간주나무 에탄올 추출물이 미치는 영향을 알아본 결과 노간주나무 에탄올 추출물은 100 ㎍/㎖에서 67.1%의 MMP-1의 발현 저해효과를 나타내었다. 노간주나무 에탄올 추출물의 MMP-1의 단백질 및 mRNA의 발현은 50 ㎍/㎖의 농도에서 각각 35%, 39%의 저해율을 나타냄에 따라 노간주나무 에탄올 추출물은 collagen을 분해하는 효소인 MMP-1의 발현을 억제하고 광노화에 의한 주름생성 억제 활성을 보이는 것을 확인하였다. 이와 같은 결과로 미루어 보아 열수 추출물보다 에탄올 추출물이 더 높은 효과를 나타냄을 확인할 수 있었으며, 노간주나무 추출물의 미백 및 주름개선 효과를 이용하여 기능성 화장품 소재로서의 가능성을 확인할 수 있었다.

This study was intended to investigate antioxidative potency, whitening, and improving wrinkles of Juniperus rigida Sieb., which is the world-wide inhabited cedar tree kind. The result of the examination measuring the polyphenol content by investigating antioxidativity of Juniperus rigida Sieb. extract showed 71.3 ㎎/g in water extract, and 116.0 ㎎/g in ethanol extract; flavonoid content in water extract was 17.7 ㎎/g, and in 76.4 ㎎/g in ethanol extract. Measuring nitrite radical scavenging activity, 1,000 ㎍/㎖ water extract and ethanol extract of Juniperus rigida Sieb. showed 44.0% and 86.8% efficacy respectively, displaying similar efficacy as control group of the study BHT; after measuring ABTS radical cation decolorization, it has shown that in 1,000 ㎍/㎖ of water extract and ethanol extract, they showed 91.4% and 91.1% of scavenging activity respectively, which demonstrate the parallel activity to the same concentration of ascorbic acid. FRAP was investigated, showing 1.83 mM efficacy in water extract and 1.77 mM in ethanol extract, higher in water extract. Then, SOD like activity in water extract and ethanol extract with the concentration level of 1,000 ㎍/㎖ showed 43.4% and 32.3%, and xanthine oxidase inhibition vitality activity in water extract and ethanol extract with the concentration level of 1,000 ㎍/㎖ showed 52.7% and 72.3% inhibition efficacy. Measuring the elastase to investigate whitening efficacy, it has shown that in the concentration level of 1,000 ㎍/㎖ of the two extracts, water extract showed lower than 10% of inhibition activity while ethanol extract showed 68.5% inhibition activity; collagenase inhibition activity in water extract and ethanol extract showed 44.9% in water extract, 97.2% in ethanol extract, which in the case of ethanol extract, it has parallel efficacy to ascorbic acid (99.6%). Moreover, measuring biosynthesis of collagen by fibroblast, it is implied that the concentration level of 50 ㎍/㎖ of ethanol extract showed 151.52% of biosynthetic promotion, proving that ethanol extract improves wrinkles superbly. The result of investigation done on influence of ethanol extract on MMP-1 caused by UVA shows that the concentration level of 100 ㎍/㎖ of ethanol extract of Juniperus rigida Sieb. has 67.1% inhibition activity to restrain MMP-1 appearance. The concentration level of 50 ㎍/㎖ ethanol extract of Juniperus rigida Sieb. showed 35% and 39% inhibition ratio to MMP-1 protein and mRNA respectively, thereby restraining appearance of collagen breakdown enzyme MMP-1 and wrinkle creation by skin photoaging. Thus, to infer from the result of the study, ethanol extract had greater efficacy than water extract and the Juniperus rigida Sieb. extracts could be utilized as materials for functional cosmetics such as whitening product or anti-wrinkle product.
