Stabilin-2 Acts as an Engulfment Receptor for the Phosphatidylserine-dependent Clearance of Primary Necrotic Cells :

Necrosis is the untimely, rapid, irreversible demise of cells triggered by trauma, bacterial or viral toxins, extreme temperature changes, nutrient deprivation or chemical agents. The elimination of necrotic cells exemplifies a greater challenge for the phagocytes due to their ruptured and leaky plasma membrane and the underlying molecular mechanism remains ambiguous. Necrotic cell death is associated with several pathologies like the neurodegenerative disorders, autoimmune diseases and ischemia, thus efficient disposal of these necrotic cells is essential to limit the spread of infection to healthy cells and tissues. Phosphatidylserine (PS) exposed by necrotic cells serves as an engulfment signal for their effective clearance. In this study, we report that stabilin-2 acts as a receptor for necrotic cell clearance. Stabilin-2-mediated necrotic cell engulfment occurred in a PS-dependent manner. EGF-like domain repeat (a PS-recognition domain of stabilin-2) bound to necrotic cells and inhibited the stabilin-2-mediated engulfment. However, the ligation of PS exposed on primary necrotic cells to Stabilin-2 was inadequate to induce the anti-inflammatory effect in stabilin-2-expressing cells, a prominent attribute of Stabilin-2 receptor. These findings facilitate the elucidation of the molecular mechanisms responsible for the PS-dependent clearance of necrotic cells.
