Fabrication of elastic hydrogels is one of the greatest challenges in tissue engineering research. Elastic hydrogel with minimal toxicity and maximum biocompatibility are considered as key issues to avoid any toxic effect and enhance feasibility for biomedical application. In this research, we select hyaluronic acid and Pluronic-F127 to synthesis elastic hydrogel. Hyaluronic acid is known as biocompatible and biodegradable polymer, degrade rapidly in human body. Pluronic-F127 also easily diffuses in vivo conditions. To overcome rapid diffusion and degradation we introduce cross-linking method. Hydroxyl group of hyaluronic acid and hydroxyl end group of PF127 was modified by vinyl group. Acrylated-hyaluronic acid(HA) and Pluronic-Diacrylated was prepared in different mixing ratio. Then acrylated-hyaluronic acid and pluronic-Diacrylate was dissolve in a solution and was cross-linked by UV-irradiation. Several characterization parameter were applied to characterized the chemical and physical properties of the HA/Pluronic hydrogel. This HA/Pluronic hydrogels shows enhanced mechanical strength, extensibility and in cell viability. Elastic hydrogel A, B, C shows enhanced strength with increasing pluronic contents from 4 to 8wt.%. Also it can shows high mechanical strength with higher UV-irradiation time. It can swell 5 ~ 20 times volumes in PBS. These hydrogels can be extensible 2 ~ 3times length. Then FE-SEM image shows compact pore size with increasing pluronic contents. Each hydrogel shows surface pore size 20~50µm and cross-section image 10 ~ 30µm. HA, A-HA, Pluronic, Plu-DA were examined in cell viability test(MTT assay). Each material shows higher cell viability than control conditions. As a result these hydrogels can be apply to anti-adhesive agent, cell laden hydrogels, drug delivery system, and tissue engineering application.
