This study was conducted to enhance dissolution rate of celecoxib and aceclofenac with extremely low solubility and high permeability (BCS class II) in water using polyvinyl pyrrolidone (PVP), hydroxyproylmethyl cellulose (HPMC) and sodium lauryl sulfate (SLS) as carriers of solid dispersion. Solid dispersions were prepared by spray drying method and rotary evaporation method as solvent evaporation methods using different ratios of drugs and polymers. The characterization of the prepared solid dispersions was characterized by scanning electron microscopy (SEM), Fourier transformation infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC) and powder x-ray diffractometer (PXRD). The dissolution behavior of the prepared solid dispersions was compared with commercial celecoxib and aceclofenac, CelebrexⓇ (Pfizer, Co, USA) and AirtalⓇ (Deawoong, Co, Korea) as control groups in simulated phosphate buffer at pH 1.2 and 6.8. The dissolution rate of the drug was affected by nature and amount of polymer used. Celecoxib, the prepared solid dispersion using carriers of both PVP and HPMC appeared to have the highest dissolution rate. In aceclofenac, the prepared solid dispersion as ratios of drug and carriers (1:5) appeared to have the highest dissolution rate. Therefore, solid dispersion techniques of spray drying and rotary evaporation methods can be successfully used to improve the dissolution rate of the water-insoluble drugs.
