Purpose : Parkinson's disease is one of the neurodegenerative disease, showing impairment of voluntary movement induced by death of dopaminergic neurons in substantia nigra and striatum. PAK1 activity is closely related with cell survival or death. In this study, alteration of PAK1 activity was revealed using Parkinson's disease model rat. In order to determine the relationship between PAK1 activity and survival of neurons were examined in the cultured dopaminergic neurons and the hemiparkinson rats, and voluntary behavior by expression of DN-PAK1. Materials and methods : First, in order to identify the correlation of 6-OHDA treatment with PAK1 activity in dopaminergic neurons, 6-OHDA was treated on the cultured dopaminergic neurons and then PAK1 activity was measured by immunocytochemistry staining. Second, rats were injected with 6-OHDA in striatum by microinfusion using stereotaxic surgery. And then, to identify the PAK1 activity, Western blotting analysis was performed using the midbrain tissue lysates. Moreover, alteration of dopaminergic neuronal mass in substantia nigra was measured by immunostaining. Finally, lentiviruses which are expressing GFP or DN-PAK1 were injected and expressed in substantia nigra of rat midbrain, and then behavioral deficit examined. Results : PAK1 activity was decreased by 6-OHDA treatment, well known to induce cell death by producing reactive oxygen species, in the cultured Mesencephalic dopaminergic neurons and the rat midbrain tissue. Next, Dopaminergic neuronal death in Substantia nigra was induced by injection of 6-OHDA. Moreover, DN-PAK1 overexpressed rats showed significantly reduced voluntary movement of contralateral side of the paw. Conclusion : Decreased PAK1 activity induces dopaminergic neuronal death. Therefore, it suggests that activity change of PAK1 may be an important regulatory mechanism in developing Parkinson's disease.
