혐기성 상태에서 암모니아성 질소는 전자공여체로, 아질산성 질소는 전자수용체로 반응하여 질소가스와 질산성 질소가 생성되는 혐기성 암모늄 산화 미생물을 이용한 질소제거 공정은 재래식 질산화/탈질소화 공정에 비해 비용이나 처리 효율면에서 아주 유리하다. 하지만 Anammox 미생물은 배가시간이 느리기 때문에 파일럿 또는 실규모 플랜트에 적용했을 때 미생물이 wash-out되는 현상이 생기면 정상적인 운전이 어렵고 회복에도 많은 시간이 소요된다는 단점이 있는데, 이러한 문제는 미생물을 고분자 담체에 포괄 고정화시킴으로써 해결할 수 있다. 본 연구에서는 미생물을 포괄 고정화시킬 고분자 담체로써 PEG (Poly Ethylene Glycol)을 선정하였다. 입상형태와 PEG Gel 고정화 혐기성 암모늄 산화균을 연속 배양하여 기질농도 변화에 따른 미생물 활성과 산소노출 유무에 따른 특성을 평가하였다. 입상형태와 PEG Gel Anammox 미생물은 각각 부피 3.3L의 아크릴 반응기에 식종하여 2 세트로 운전하였고 고농도 질소를 함유한 합성폐수를 제조하여 유입수로 하였다. 유입수와 유출수에 대해 NH4+-N, NO2--N, NO3--N, 알칼리도, 온도, pH를 분석하여 미생물의 활성을 평가하였다. 먼저 입상형태 미생물은 유입수의 기질농도 변화 실험에서 T-N농도를 40mg/L에서 500mg/L까지 단계적으로 높여주었을 때 NH4+-N, NO2--N, T-N의 평균 제거율은 90.5±6.5, 96.6±4.9, 93.2±6.1%로써 안정적인 반응을 보였으며, PEG Gel 미생물은 유입수의 T-N농도를 10mg/L에서 500mg/L까지 증가시켰을 때 각각 83.3±13.0, 96.4±6.1, 90.3±7.5%의 제거율을 보여주었다. PEG Gel과 입상형태 모두 유입되는 T-N의 농도가 높을수록 제거율이 감소하는 추세를 보였고, 각 유입수의 질소농도에서는 입상형태의 Anammox 미생물이 PEG Gel보다 제거율이 더 높게 나타났다. 다음으로는 반응기 운전기간 244일 중 100일까지는 유입 합성폐수에 질소가스로 탈기하지 않고 미생물이 산소에 노출되도록 조정하였고 그 이후에는 질소가스를 탈기하여 유입수의 DO농도를 낮추어 주는 조건으로 산소노출 유무에 따른 미생물의 활성을 조사하였다. 입상형태의 미생물은 유입수의 DO농도가 8.4±0.23mg/L였을 때 암모니아성 질소, 아질산성 질소, 총 질소의 제거율은 81.3±3.84, 98.9±1.20, 90.6±2.33%였고 DO농도가 0.7±0.11mg/L일 때는 85.8±3.83, 99.2±0.32, 92.7±1.82%로써 산소에 노출되지 않았을 때 질소 제거율이 소폭 증가하였으나 큰 차이가 없는 것으로 보아 이미 활성을 띈 입상형태의 미생물은 산소노출 유무에 영향을 받지 않는 것으로 판단된다. PEG Gel 미생물의 경우, 탈기 전의 암모니아성 질소, 아질산성 질소, 총 질소의 제거율은 58.1±13.32, 90.7±8.66, 76.1±7.92%였고 질소가스로 탈기했을 때의 제거율은 72.1±11.36, 98.0±1.13, 85.6±5.57%로 나타났다. 즉, 산소에 노출되지 않았을 때가 노출되었을 때 보다 질소 제거율이 더 높고 표준편차가 적은 것으로 보아 PEG Gel Anammox가 입상형태의 Anammox보다 산소노출에 더 민감하게 반응하는 것으로 판단된다.

But when anaerobic ammonium oxidation bacteria is washed out in the pilot or full scale plant, it is very hard to recover normal condition and needs lots of time to get back because anaerobic ammonium oxidation bacteria has long doubling time. These problems can be solved by whole cell entrapment of anammox bacteria. In this study, PEG(Poly Ethylene Glycol) gel was selected as a media in the whole cell entrapment. The activity of anaerobic ammonium oxidizer with synthetic media and granular form was studied in the same two reactors to investigate the effect of influent nitrogen concentration and exposure of oxygen. Granular Anammox and PEG Gel Anammox were seeded into acrylic reactor and synthetic wastewater including high concentration of nitrogen was introduced to the reactors. The activity of anammox bacteria was estimated by analyzing ammonium, nitrite, nitrate, alkalinity, temperature and pH. First of all, when concentration of influent total nitrogen was changed from 40mg/L to 500mg/L, average removal efficiency of ammonium, nitrite and nitrate were shown to 90.5±6.5, 96.6±4.9, 93.2±6.1%, respectively. And, concentration of influent total nitrogen was changed from 10mg/L to 500mg/L in the PEG Gel Anammox test, average removal efficiency of ammonium, nitrite and nitrate were shown to 83.3±13.0, 96.4±6.1, 90.3±7.5%, respectively. The activity of granular Anammox showed a better performances than that of PEG Gel Anammox in the first test. In second test, from 1 to 100days of total operation period, anaerobic ammonium oxidizer was exposed to oxygen and after that nitrogen gas was sparged to influent synthetic wastewater for the removal of residual dissolved oxygen in the influent. In the granular Anammox reactor, removal efficiency of ammonium, nitrite and nitrate at 8.4±0.23mg/L of DO concentration showed 81.3±3.84, 98.9±1.20, 90.6±2.33%, respectively. And when DO concentration was controlled to 0.7±0.11mg/L, removal efficiency of ammonium, nitrite and nitrate showed 85.8±3.83, 99.2±0.32, 92.7±1.82%, respectively. It suggest that the activated anaerobic ammonium oxidizer is not highly affected to exposure of oxygen in the influent.
