이 논문은 야파광의 질, 질 변환 및 식물체 조사부위에 따른 3종의 모델식물인 페튜니아(질적 장일식물), 제라니움(중일식물) 및 국화(질적 단일식물)의 형태형성, 개화, 그리고 광수용체 관련 유전자의 전사에 관해 연구하였다. 또한 청색광을 보광 또는 야파광으로 공급시 국화의 형태형성, 개화, 그리고 광수용체 관련 유전자의 전사에 대해서도 알아보았다. 명기에는 백색광(white LEDs, 400-700 nm)으로 180 μmol•m-2•s-1 PPFD의 광도를 공급하였다. 장일[LD, 16 h/8 h(명기/암기)], 단일[SD, 10 h/14 h(명기/암기)] 및 단일조건의 암기에 10 μmol•m-2•s-1 PPFD의 광도로 4시간 동안 조사한 단일-야파(NI)의 세 가지 일장조건을 처리하였다. 모델식물의 형태형성과 개화에 미치는 야파광 질의 영향을 보기 위한 실험 1에서는 청색(NI-B, 450 nm), 녹색(NI-G, 530 nm), 적색(NI-R, 660 nm), 근적색(NI-Fr, 730 nm) 및 백색(NI-W, 400-700 nm)을 야파광질로 사용하였다. 장일식물인 페튜니아의 신초장과 중일식물인 제라니움의 초장은 NI-Fr에서, 단일식물인 국화의 초장은 LD에서 가장 컸다. 장일식물은 LD, NI-G, NI-R, NI-Fr, NI-W에서 개화하였다. 중일식물은 모든 처리에서 개화하였으며 야파광의 질에 따라 차이가 없었다. 단일식물은 SD, NI-B 및 NI-Fr에서 개화하였다. 장일식물의 야파처리에서는 광수용체 관련 유전자의 발현이 전반적으로 높았으며, 특히 phyA(phytochrome A), phyB(phytochrome A) 및 cry1(cryptochrome 1)의 발현이 NI-G, NI-B, NI-R에서 가장 높았다. 중일식물은 LD에서 phyB와 FTL(FLOWERING LOCUS T)를 제외한 모든 유전자의 발현이 높았다. 단일식물은 NI-B, NI-Fr, SD에서 phyA와 cry1의 발현이 높았다. 따라서 실험에 사용된 모든 일장반응식물의 형태형성, 개화 및 광수용체 관련 유전자의 발현은 야파광의 질에 영향을 많이 받았고, 특히 단일식물에서 그 영향이 컸다. 또한 단일조건 하에서 NI-W와 NI-R로 야파시 장일식물의 형태형성과 개화를 조절할 수 있었다. 실험 2는 야파광의 질 변환에 따른 모델식물의 형태형성과 개화에 관한 내용으로 청색, 적색, 근적색 및 백색 야파광을 조사개시 2시간후에 변환하여 조사하였다. 대조구로 장일과 단일을 두고 총 12처리의 야파광 조합[청색→적색(NI-BR), 적색→청색(NI-RB), 적색→근적색(NI-RFr), 근적색→적색(NI-FrR), 청색→근적색(NI-BFr), 근적색→청색(NI-FrB), 백색→청색(NI-WB), 청색→백색(NI-BW), 근적색→백색(NI-FrW), 백색→근적색(NI-WFr), 적색→백색(NI-RW), 백색→적색(NI-WR)]을 처리하였다. 장일식물의 신초장은 NI-RFr에서 가장 컸고, NI-RW와 NI-WR에서 가장 작았다. 중일식물과 단일식물의 초장은 Fr이 포함된 야파처리에서 컸다. 장일식물은 LD, NI-BR, NI-RB, NI-RFr, NI-WB, NI-BW, NI-RW, NI-WR에서 개화하였다. 중일식물은 모든 처리에서 개화하였고 Fr이 포함된 야파처리에서 개화가 촉진되었다. 단일식물은 청색광과 적색광의 조합인 NI-RB처리, 청색광과 백색광의 조합인 NI-WB처리에서 개화하였다. 실험한 모든 모델식물의 phyA, phyB, cry1 발현은 NI-RFr과 NI-FrR에서 높았다. 장일, 단일 및 단일-야파와 같은 광주기는 모든 일장반응식물의 형태형성과 개화에 영향을 미쳤다. 하지만 모든 모델식물의 형태형성과 개화에 있어서 첫 번째 야파광은 영향이 적거나 없었고 두 번째 야파광의 영향이 컸다. 실험 3은 야파광의 식물체 조사부위와 야파광질에 따른 모델식물의 형태형성과 개화에 관한 내용으로 청색(NI-B), 적색(NI-R), 근적색(NI-Fr), 백색(NI-W)광을 정단(ST), 어린 잎(YL) 또는 늙은 잎(OL)에 조사하였다. 장일식물의 신초장과 단일식물의 초장은 ST, YL 및 OL에 NI-Fr을 조사하였을 때 가장 컸으나, 중일식물의 초장은 ST에 NI-R을 조사하였을 때 가장 컸다. 장일식물은 YL에 NI-R, OL에 NI-W를 조사한 경우 및 LD에서 개화하였다. 중일식물은 모든 처리에서 개화하였다. 단일식물의 ST, YL 및 OL에 NI-B, NI-Fr, NI-W를 조사한 경우 및 SD에서 개화하였다. 하지만 단일식물의 경우NI-R을 YL이나 OL에 조사시에는 개화하였으나 ST에 조사시에는 개화하지 않았다. 모든 모델식물의 ST에 NI-B를 야파하였을 때 phyA, phyB, cry1, FTL 및 AFT(Anti-florigenic FT/TFL1 familyprotein)의 발현이 가장 높았지만, 다른 조사부위에서는 차이가 없었다. 단일조건 하에서 YL에 NI-R을 조사하거나 OL에 NI-W를 조사하면 장일식물의 개화를 유도할 수 있을 것이다. 또한 OL에 NI-B를 조사하면 기술은 초장을 억제시켜 분화용 단일식물 생산에 적용 가능성이 있다. 하지만 실용적인 화훼작물의 개화조절을 위해서는 야파기술에 대한 심도 있는 추가연구가 필요하다. 실험 4는 청색광의 보광과 야파광에 따른 국화의 형태형성과 개화에 관한 내용인데, 1) 청색광 없이 명기만 10시간(SD10) 처리한 대조구, 2) 9시간 명기에 연속한 4시간 청색광 처리(SD9 + 4B), 3) 13시간 명기에 4시간 청색광 야파처리(LD13 + NI-4B), 4) 10시간 명기에 4시간 청색광 야파처리(SD10 + NI-4B) 및 5) 13시간 명기에 연속한 4시간 청색광 처리(LD13 +4B)를 두었다. 초장은 LD13 + NI-4B에서 가장 컸다. 모든 처리에서 개화하였지만 LD13 + 4B에서는 LD13 + NI-4B에 비해 개화가 촉진되었고 대조구에 비해 식물체당 개화수도 많았다. 청색광은 장일조건에서도 단일식물인 국화의 개화와 화아형성을 촉진시켰기에, 이것을 이용한 개화조절은 암막을 대체할 수 있는 가능성이 있지만 실용화를 위해서는 청색광의 보광에 관한 추가연구가 필요하다.

Effect of the quality, quality shifting, and positioning of night interruption light (NIL) on morphogenesis, flowering, and transcription of photoreceptor genes was investigated in Petunia hybrida Hort. ‘Easy Wave Pink’ (qualitative long day plant, LDP), Pelargonium × hortorum L.H. Bailey ‘Ringo 2000 Violet’ (day neutral plant, DNP), and Dendranthema grandiflorum ‘Gaya Yellow’ (qualitative short day plant, SDP). Effect of supplementary and night interruptional blue (B) light on morphogenesis, flowering, and transcription of photoreceptor genes of Dendranthema grandiflorum ‘Gaya Yellow’ (qualitative SDP) was also examined. Plants were grown in a closed-type plant factory under a light intensity of 180 μmol•m-2•s-1 PPFD provided by white (W) light emitting diodes (LEDs) under a condition of either long day (LD, 16 h light/8 h dark), short day (SD, 10 h light/14 h dark), or SD with 4 h night interruption (NI) condition. The NI was provided by 10 ± 3 μmol•m-2•s-1 PPFD. In the first experiment, the NIL was provided by blue (NI-B), green (NI-G), red (NI-R), far-red (NI-Fr), or white (NI-W) LEDs. Shoot length or plant height of the LDP (petunia) and DNP (geranium) was the greatest in the NI-Fr, while that of the SDP (chrysanthemum) was the greatest in the LD. In the LDP, flowering was observed in the LD, NI-G, NI-Fr, and NI-W. Flowering of the DNP was not affected by quality of the NIL, and all plants flowered in all treatments. Flowering of the SDP was observed in the SD, NI-B, and NI-Fr. In the LDP, regulation in the expression of the photoreceptor genes was observed by the NI, and especially there was a tendency that the phyA, phyB, and cry1 were highly expressed in the NI-G, NI-B, and NI-R as compared to the LD and SD. Under normal the LD all photoreceptor genes except the phyB and FTL in the DNP were well expressed. In the SDP, the phyA and cry1 were highly expressed in the NI-B, NI-Fr, and SD. These results suggest that morphogenesis, flowering, and transcriptional factors of these plants were highly affected by quality of the NIL, especially in the SDP. Furthermore, the NI-R or NI-W was the most suitable in the NI strategy of controlling morphogenesis and flowering of the LDPs during SD seasons. In the second experiment, the quality of NIL was shifted from one to another after first 2 h in sets of two qualities among B, R, Fr, and W. The LD and SD were used as the control, and 12 SD treatments with shifting of NIL quality by LEDs were as follows: from blue to red (NI-BR), from red to blue (NI-RB), from red to far-red (NI-RFr), from far-red to red (NI-FrR), from blue to far-red (NI-BFr), from far-red to blue (NI-FrB), from white to blue (NI-WB), from blue to white (NI-BW), from far-red to white (NI-FrW), from white to far-red (NI-WFr), from red to white (NI-RW), and from white to red (NI-WR). Shoot length of the LDP (petunia) was the greatest in the NI-RFr and the least in the NI-RW and NI-WR. Plant height of the DNP (geranium) and SDP (chrysanthemum) was greater in those NI treatments consisted of the Fr as compared with other treatments, and it was the least in the NI-WB among the NI treatments. Flowering of the LDP was observed in the LD, NI-BR, NI-RB, NI-RFr, NI-WB, NI-BW, NI-RW, and NI-WR. Flowering of the DNP was observed in all treatments, and especially was promoted by the Fr-containing NIL regardless of the order of light quality given. Flowering of the SDP was observed in the NI-RB, NI-FrR, NI-BFr, NI-FrB, NI-WB, NI-FrW, NI-WFr, NI-WR and SD of course, and especially was promoted in the NI-BFr and NI-FrB. The phyA, phyB, and cry1 genes were highly expressed in the NI-RFr and NI-FrR regardless of photoperiodic response group of the plant, i.e. in all LDP, DNP, and SDP. Photoperiod affected both morphogenesis and flowering in the LDP, DNP, and SDP. While the first NIL did affect neither morphogenesis nor flowering, the second NIL significantly affected not only morphogenesis but also flowering, in all plants. In the third experiment, the NIL was provided by B (NI-B), R (NI-R), Fr (NI-Fr), or W (NI-W) LEDs. The NIL was positioned to irradiate the shoot tip (ST), youngest mature leaf (YL), or old leaf (OL). Shoot length and plant height were the greatest in the NI-Fr onto the ST, YL or OL in the LDP (petunia) and SDP (chrysanthemum), and were the greatest in the NI-R onto the ST in the DNP (geranium). Leaf growth was severely affected when the NIL was targeted onto the ST in all plants. Flowering of the LDP was observed in the LD, NI-R onto the YL, and NI-W onto the OL. Flowering of the DNP was induced in all treatments. Flowering of the SDP was observed in the SD, NI-B, NI-Fr, and NI-W onto the ST, YL or OL. Flowering of the SDP occurred in the NI-R onto the YL or OL, but not the NI-R onto the ST. The phyA, phyB, cry1, FTL, and AFT were highly expressed in the NI-B onto the ST, whereas the expression of these genes was not changed in all other treatments. The NI-R or NI-W onto the YL or OL could potentially be used as a tool for the flowering control light in the LDPs during SD seasons. The NI-B onto the OL has a potential applicability to use in production of potted SDPs due to subsequent height inhibition. Further studies are needed to investigate the effect of the B light supplementation on flowering control of the plant. In the fourth experiment, the control plant was exposed to 10 h short day treatment (SD10) without the B light. The B light was used for 4 h either 1) to extend the photoperiod at the end of 9 h SD as a sole light source (SD9 + 4B), 2) for the NI in the 13 h long day (LD13 + NI-4B), 3) for the NI in the 10 h SD (SD10 + NI-4B), or 4) to supplement the W LEDs at the end of a 13 h LD (LD13 + 4B). The supplementary or night interruptional blue (B) light was provided by 10 μmol•m-2•s-1 PPFD using the B LEDs. Plant height was the greatest in the LD13 + NI-4B. Flowering was induced by all treatments, and the days after treatment initiation to visible flower bud or days to visible bud (DVB) was shortened in the LD13 + 4B as compared to the LD13 + NI-4B. Number of flowers per plant increased in the LD13 + 4B as compared to the SD10. The expression of phyA, phyB, and cry1 was very high in the SD9 + 4B, LD13 + 4B, LD13 + NI-4B, and SD10 + NI-4B as compared to the SD10. The results indicate that the B light used as a supplementary light for 4 h during the photoperiod promotes flowering and number of flower buds formed. The results present a useful practical technique for flowering control as an alternative method of using the blackout curtains during the LD seasons. However, further studies on the application methods of the B light for control of flowering of the SDPs are necessary.

Plant height of the DNP (geranium) and SDP (chrysanthemum) was greater in those NI treatments consisted of the Fr as compared with other treatments, and it was the least in the NI-WB among the NI treatments. Flowering of the LDP was observed in the LD, NI-BR, NI-RB, NI-RFr, NI-WB, NI-BW, NI-RW, and NI-WR. Flowering of the DNP was observed in all treatments, and especially was promoted by the Fr-containing NIL regardless of the order of light quality given. Flowering of the SDP was observed in the NI-RB, NI-FrR, NI-BFr, NI-FrB, NI-WB, NI-FrW, NI-WFr, NI-WR and SD of course, and especially was promoted in the NI-BFr and NI-FrB. The phyA, phyB, and cry1 genes were highly expressed in the NI-RFr and NI-FrR regardless of photoperiodic response group of the plant, i.e. in all LDP, DNP, and SDP. Photoperiod affected both morphogenesis and flowering in the LDP, DNP, and SDP. While the first NIL did affect neither morphogenesis nor flowering, the second NIL significantly affected not only morphogenesis but also flowering, in all plants. In the third experiment, the NIL was provided by B (NI-B), R (NI-R), Fr (NI-Fr), or W (NI-W) LEDs. The NIL was positioned to irradiate the shoot tip (ST), youngest mature leaf (YL), or old leaf (OL). Shoot length and plant height were the greatest in the NI-Fr onto the ST, YL or OL in the LDP (petunia) and SDP (chrysanthemum), and were the greatest in the NI-R onto the ST in the DNP (geranium). Leaf growth was severely affected when the NIL was targeted onto the ST in all plants. Flowering of the LDP was observed in the LD, NI-R onto the YL, and NI-W onto the OL. Flowering of the DNP was induced in all treatments. Flowering of the SDP was observed in the SD, NI-B, NI-Fr, and NI-W onto the ST, YL or OL. Flowering of the SDP occurred in the NI-R onto the YL or OL, but not the NI-R onto the ST. The phyA, phyB, cry1, FTL, and AFT were highly expressed in the NI-B onto the ST, whereas the expression of these genes was not changed in all other treatments. The NI-R or NI-W onto the YL or OL could potentially be used as a tool for the flowering control light in the LDPs during SD seasons. The NI-B onto the OL has a potential applicability to use in production of potted SDPs due to subsequent height inhibition. Further studies are needed to investigate the effect of the B light supplementation on flowering control of the plant. In the fourth experiment, the control plant was exposed to 10 h short day treatment (SD10) without the B light. The B light was used for 4 h either 1) to extend the photoperiod at the end of 9 h SD as a sole light source (SD9 + 4B), 2) for the NI in the 13 h long day (LD13 + NI-4B), 3) for the NI in the 10 h SD (SD10 + NI-4B), or 4) to supplement the W LEDs at the end of a 13 h LD (LD13 + 4B). The supplementary or night interruptional blue (B) light was provided by 10 μmol•m-2•s-1 PPFD using the B LEDs. Plant height was the greatest in the LD13 + NI-4B. Flowering was induced by all treatments, and the days after treatment initiation to visible flower bud or days to visible bud (DVB) was shortened in the LD13 + 4B as compared to the LD13 + NI-4B. Number of flowers per plant increased in the LD13 + 4B as compared to the SD10. The expression of phyA, phyB, and cry1 was very high in the SD9 + 4B, LD13 + 4B, LD13 + NI-4B, and SD10 + NI-4B as compared to the SD10. The results indicate that the B light used as a supplementary light for 4 h during the photoperiod promotes flowering and number of flower buds formed. The results present a useful practical technique for flowering control as an alternative method of using the blackout curtains during the LD seasons. However, further studies on the application methods of the B light for control of flowering of the SDPs are necessary.
