The present study aimed to evaluate potential alleviation effects of HJ on high fat diet (HFD) induced obesity and its related complications by 3T3-L1 cell and C57BL/6 mice. We carried out HFD for 12-weeks and administered orally HJ for 8-weeks in a dose dependent manner in C57BL/6 mice. At the end of experiment, blood was prepared for biochemical analysis and leptin ELISA assay. Epididymal, mesenteric fat and liver were removed surgically, weighed and observed by histological or immunohistochemical changes. 3T3-L1 adipocyte cells were cultured, assessed lipid accumulation by Oil Red O staining and expression of PPARγ and C/EBPα by western blotting. HJ significantly prevented body weight, lipid accumulation and alleviated hyperlipidemia by decreasing levels of triglyceride (TG) and low density lipoprotein (LDL), and the atherogenic index in serum. Furthermore, HJ decreased elevated serum leptin and subcutaneous adipocyte sizes. Elevated serum levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in HFD fed mice were decreased in HJ treatment group. HJ treatment significantly decreased HFD-induced liver weight and lipid accumulation in the liver. Oil Red O staining region, PPARγ and C/EBPα expressions in adipocyte were suppressed by HJ treatment in the 3T3-L1 adipocte cells. HJ treatment alleviated HFD-induced obese status and obesity related complications such as hyperlipidemic change that induce cardiovascular disease and NAFLD.
