We isolated full length cDNA of these four genes and its corresponding genomic clone containing both exon and intron and compared them among the three taxa mention above; this comparison revealed that there was no sequence variation. To check if differential expression of these key genes resulted in difference in leaf phenotype, transcriptome analysis by quantitative Real-Time PCR and also RNA in situ hybridization was carried out. The RT PCR result of leaves regenerated from var. chaerophylloides petiole explant showed elevated levels of VaKN1 and VaSTM gene expression in the compound leaves which were morphologically similar to those of var. takahashii and var. chaerophylloides in relative to var. albida. VaCUC2 expression was found to be increased in leaves that were similar to that of var. chaerophylloides and var. takahashii. But when the expression level was checked the compound leaved plants of var. chaerophylloides and var. takahashii showed elevated expression of VaKN1 gene in the leaves. This result also correspond with the results of RNA in situ hybridization; no expression of VaKN1 and VaSTM genes were found outside the shoot apical meristem (SAM) in the simple leaved var. albida, whereas in the case of the compound leaved var. takahashii and var. chaerophylloides VaKN1 expression was detected outside the SAM. In var. chaerophylloides, VaKN1 expression was detected at the margins of the leaves, suggesting that they maintain an indeterminate fate for a while at the margin leading to leaflet generation and thereby leading to compound leaves.
