Functional single-wall carbon nanotubes (SWNTs) have been proposed and actively explored as multipurpose, innovative medical materials for applications in fields such as materials science, drug delivery, and diagnostics. However, the nature of SWNT interactions and their toxicity are not clearly understood. Because of the extensive potential commercial application of SWNTs, their health risk assessment is critical. Recent studies have shown that nanoparticles may be a valuable tool in stroke prevention and treatment. However, the effects of carbon nanotubes (CNTs) themselves used as a drug rather than as a medical substrate have been less investigated. In this research, the solubility and safety of polyethylene glycol-functionalized (PEG)-SWNTs was first examined to highlight the features a molecule must possess to safely function in a water-based body. No change was detected in the activity of two liver enzymes used as markers of hepatotoxicity (glutamic oxaloacetic transaminase/glutamic pyruvic transaminase) or in the morphology of the liver, spleen, or kidney in PEG-SWNT-administered rats compared with PBS-injected controls. When SWNTs were administered to rats pre- or post-middle cerebral artery occlusion (MCAO), a rat model of ischemic stroke, infarct volumes and the apoptotic cell number were significantly reduced and neurological scores on the rotarod treadmill test were increased at 1-week post reperfusion. These improvements were associated with a decrease in the expression of the anti-apoptotic protein Bcl-2, and an increase in the activation of extracellular signal-regulated kinase and Akt in brain tissues. Treatment with SWNTs either pre- or post-MCAO reduced the number of GFAP-positive and Iba-1-positive cells in brain tissues. Additionally, Fourier transform-Raman spectroscopy revealed that systemically administered SWNTs were detectable and stable in rat brains for 1 week. In conclusion, these findings provide evidence that SWNTs are safe and protect neurons against ischemic injury and repair brain damage, and they suggest that SWNT are an excellent candidate for repurposing as a nano-drug for stroke prevention and treatment.
