Purpose : Parkinson's disease (PD) is a neurodegenerative disease, caused by the death of dopaminergic neurons in the substantia nigra, a region of the midbrain. To identify the role of PAK1 in pathogenesis of PD, the effect of activated PAK1 on death of tyrosine hydroxylase (TH) positive cell and behavioral changes was examined in the rat models of PD. Matherials and Methods : To develop PD animal models, 6-OHDA or MPTP was infused to the striatum region of rats by microfusion using stereotaxic surgery. Simultaneously, PAK1-CA or GFP lentivirus was injected to the substantia nigra. To measure behavioral defects, forearm usage test was performed. The level of dopaminergic (DA) neurons in the substantia nigra was analyzed by an immunostaining to TH. In addition, to determine the effects of Cyclosporin A on neuronal death by 6-OHDA or MPTP treatments, PD animals was treated with Cyclosporin A by intraperitoneal injection and subjected to behavioral assay and immunostaining. Results : PAK1-CA expression rescued loss of dopaminergic neurons in the substantia nigra and behavioral defects induced by 6-OHDA or MPTP treatments. Cyclosporin A did not have significant effect in DA neuron survival, but reduced behavioral defects induced by 6-OHDA treatment. Conclusion : PAK1 activity recover loss of dopaminergic neurons by 6-OHDA or MPTP treatments. PAK1 maybe a target to recover the loss of DA neurons in PD pathogenesis.
