능동적 표적지향 약물전달 시스템은 암세포에 과발현된 특정 단백질과 특이적으로 결합하는 펩타이드, 리간드나 항체를 약물전달체의 표면에 결합하여 암세포에 선택적으로 약물을 전달하여 부작용을 최소화하기 때문에 다양하게 연구되고 있다. 본 연구에서는 약물전달체로 온도 및 외부 자기장에 감응하는 하이드로젤 나노입자를 제조하였고 입자의 표면에 타겟팅 리간드를 결합하여 표적지향성을 부여하였다. 온도 및 외부 자기장에 감응하는 하이드로젤 나노입자를 제조하기 위해 온도감응성 고분자인 N-이소프로필아크릴아미드(NIPAm)와 산화철 나노입자를 혼합하여 침전중합법으로 가교시켰다. 자궁경부암 세포인 HeLa 세포에 특이적 결합을 하는 엽산을 결합하기 위해서 하이드로젤 나노입자의 표면을 코어/쉘 형태로 제조하여 아민기로 개질하였다.
엽산이 결합된 코어/쉘 나노입자에 모델 약물인 덱사메타손을 탑재하여 12시간 이상 방출 거동을 확인하였고, 세포생존율을 측정하여 엽산이 결합된 코어/쉘 나노입자 자체가 세포의 생존율에 영향을 미치지 않는 것을 확인하였다. 입자의 표적지향 효과를 확인하기 위해서 엽산의 유무에 따른 세포내 전달 실험을 통해 엽산이 결합된 코어/쉘 나노입자와 HeLa 세포의 친화성 및 특이성을 확인하였다.

The active targeting delivery system has been studied in cancer therapy by targeting ligands to interaction between drug carriers and specific receptors on cancer cells. Even though development of external stimuli-responsive drug delivery system are essential for the sustained release drug delivery system that can minimize the side effects of the drug.
In this work, we have prepared folic acid conjugated poly(N-isopropylacylamide) (PNIPAm) microgels containing iron oxide nanoparticles for uterine cancer cells (HeLa) targeting by precipitation polymerization. To conjugate folic acid with microgels, the PNIPAm were copolymerized with amine-functionalized monomers, which can be reacted with carboxyl group of the folic acid via EDC/NHS reaction. Folic acid conjugation of microgels was confirmed with zeta potential measurement by observing shift of the isoelectric point. The biocompatibility of this folic acid conjugated microgels were studied by MTT assay. To study the effect of target specific ligands, cellular uptake of the folic acid conjugated microgels in HeLa cells was performed.
