Staphylococcus aureus continues to be a major cause of health care-related and community-associated infections. Many S. aureus infection are related to the formation of biofilm, which cause antibiotic resistance and often chronic infection. In this study, we analyzed the anti-biofilm activities of antimicrobial peptides (HG1, IB-367, and MSI-78) and antibiotics (vancomycin and ciprofloxasin) against S. aureus and methicilin-resistant S. aureus (MRSA) biofilm. Additionally we studied on the effect of extracellular DNA (eDNA) against anti-biofilm activity of the antimicrobial peptides. The antibiotics showed the bactericidal activity against planktonic cells but not against biofilm cells. On the other hand, the tested antimicrobial peptides showed the bactericidal activity against both planktonic cells and biofilm cells. Among the tested antimicrobial peptides, HG1 showed the highest bactericidal activity against biofilm cells. But as the biofilm mass of S. aureus and MRSA increased, the anti-biofilm activity of the antimicrobial peptides containing HG1 was diminished. The reason was caused by the growth of eDNA that induce resistance of biofilm cells to the antimicrobial peptides in S. aureus and MRSA biofilm. Nevertheless, HG1 showed strong anti-biofilm activity against S. auerus and MRSA biofilm. Our results suggest that HG1 can be developed as new antibiotics for the treatment of S. aureus infection.
