Sometimes, Avian coccidiosis causes concurrent infection of immuno-suppressive diseases (Gumboro, reovirus, Marek's disease, etc.) and bacterial diseases, which incur serious damage to poultry farming around the globe. In the nation, the use of foreign vaccine and addition of anticoccidial drug for prevention of coccidiosis and its treatment are arousing economic damage. Moreover, in 2020, the use of anticoccidial drug will be under overall prohibition in the nation. However, a live vaccine against coccidiosis has not been developed as yet in the nation, and thus, now is the urgent time for us to develop a domestic live vaccine against coccidiosis. In order to use a live vaccine against coccidiosis, there is the need for Eimeria which is purely isolated in one sort. It's because individual Eimeria species don't form cross-immunity with each other. Generally, a chicken is infected with more than 2 species of Eimeria, so it's not easy to secure one sort of Eimeria. That's why this study did individual isolation of Eimeria using the micro manipulation of the microscope after doing limiting dilution of Eimeria in a bid to secure one sort of isolated Eimeria. This simple method is fast and reliable, and this method makes it possible to directly isolate the individually multiplied oocyst from proliferation of Eimeria species, or the samples polluted by other types of parasite. This study isolated 4 types of Eimeria oocyst, i.e. E. acervulina, E. tenella, E. maxima, E. necatrix ,whose pathogenicity was the biggest, among 9 types of Eimeria; in addition, this study, in order to manufacture a live vaccine, amplified them individually by doing subculture of them into a chicken, and put the amplified 4 types of Eimeria oocyst in frozen storage lest other Eimeria should be contaminated. Then, this study manufactured the amplified Eimeria oocysts into a live vaccine using 2.5% Potassium dichromate, and innoculated 10-day-old chickens with the manufactured vaccine to confirm its protective immunity effect test, rate of gain, survival rate, and intestinal lesion rate. As a result, it showed 87.5% protective immunity effect, and this study was able to obtain the similar results to the non-inoculation control group in case of the rate of gain, survival rate, and intestinal lesion rate. Accordingly, it can be safely said that this study developed the method of Eimeria isolation in order to manufacture a live vaccine against Eimeria, constructed effectiveness evaluation by manufacturing a live vaccine that would replace an anticoccidial drug, and arranged the basis for substitution for an anticoccidial drug for the time to come.
