For the taxonomy studies of genus Coreoleuciscus species, measurements, meristic counts and morphometric character and phylogenetic analysis were conducted, and DNA contents analysis and interspecies hybridization experiments were carried out. In addition, the population structure analysis by CO1 gene and microsatellite marker, the origin of genus Coreoleuciscus of the river basins in the east coast, and genetic variation analysis of natural hybrid population at Hwang-ji stream (Nakdong river) were accomplished. Chapter 1. Taxonomy study of genus Coreoleuciscus species The two species are futher distinguished by the shape and/or presence of black bands on dorsal, anal and caudal fin rays. Dorsal fin of C. splendidus with a thick and discontinuous distal band and a thick and continuous basal band (versus C. aeruginos with a thin and continuous band both distally and basally). C. splendidus a has anal fin of quite thick and long distal dark band and short basal band (versus C. aeruginos has anal fin of thin and relatively short distal band and no basal band). Between the two species, there are multiple notable differences in caudal fin. C. splendidus with a total of seven bands in centrosymmetric structure; three pairs of six thin bands separated into two lobes at medial and distal-most areas and a thin basal band extending outwards. In contrast, C. aeruginos with three stripes in centrosymmetric structure; a thick and continuously band distally across lobes, one pair of two thick bands separated into two lobes medially, and a thick and short one at the basal part. Coreoleuciscus aeruginos is distinguished from C. splendidus using body coloration. C. splendidus has a golden line immediately above a luteous line that runs along a lateral line on side of body and C. aeruginos has a bluish-green line above the luteous line. During the spawning season, C. splendidus has a bright yellow line immediately above a violet and bluish line running along lateral line on side body while C. aeruginos has a luteous line immediately above a black line. Phylogenetic analysis inferred from CO1 and RAG1 gene sequence data revealed that Coreoleuciscus populations from South Korea form two independent lineages, the C. splendidus lineage comprising the Han/Geum river populations (West Korea subdistrict), and the C. aeruginos lineage comprising the Seomjin/Nakdong river populations (South Korea Subdistrict) and each lineage was supported by the highest statistical support. Distinct differences in meristic counts and measurements were not identified between C. splendidus and C. aeruginos. In contrast, this study found a high degree of morphological differentiation between the two species in body coloration, nuptial coloration, and shapes of black stripes on the dorsal, anal, and caudal fin rays. Genetic population structure of genus Coreoleuciscus species Sequence analysis of the C. splendidus Han River and Geum River populations revealed that the CO1 gene showed the FST value to range from 0.7817-1.000 (P<0.005). Ten polymorphic microsatellite DNA loci were characterized un 160 individuals from the eight populations. The average number of allele range from 6.3 to 11.4, and the average observed and expected heterozygosities ranged from 0.572-0.831 and from 0.610 to 0.857, respectively. Eight populations of C. splendidus were not discovered to the population bottleneck. Analysis of the genetic structure based on genetic distance and differentiation were discovered the significant differences between Han River and Geum River populations. Sequence analysis of the C. aeruginos Han River and Geum River populations revealed that the mitochondrial CO1 gene showed the FST value to range from 0.9567-0.9909 (P<0.005). The average haplotype and nucleotide diversities to range from 0 to 0.6474 and 0 to 0.0005, respectively. Ten polymorphic microsatellite DNA loci were characterized un 160 individuals from the eight populations. The microdistribution and distributional aspect of C. splendidus and C. aeruginos from the river basins on the east coast from 2009 to 2015. This specis was found in four independent streams of the east coast of the south Korea. C. splendidus were microdistristrubution of the east coast at Yeongok stream, Gangneungnamdae Stream, Samcheok-osip stream, and C. aeruginos was found in only one habitat at yeongdeok-osip stream. However, were not found in seven streams from Baebong, Ssang, Yangyangnamdae. Jusu, Maeup, Gagok, Uljin-wangpi streams. This species distributional zone was in the middle or upper riffles of main stream and tributaries. A natural interspecific hybrids between two Coreoleuciscus species C. splendidus and C. aeruginos was collected in their sympatric area, Hwangji stream, the Nakdong river. Morphological characters as well as RAG1 gene and CO1 gene were investigated to confirm the natural hybrid origin. Both the induced hybrids and their reciprocal counterparts displayed the double peaks of sequence chromatograms at the 13 diagnostic positions. In analysis of CO1 gene, it revealed that the maternal species of the natural hybrid appeared to be C. splendidus and C. aeruginos due to their 100% sequence identity. Thus, such result gave convincing evidence for the occurrence of a natural interspecies hybrid between C. splendidus and C. aeruginos. Therefore, the C. speldndisus populations have been introduced in the Nakdong river at recent years.

This species distributional zone was in the middle or upper riffles of main stream and tributaries. A natural interspecific hybrids between two Coreoleuciscus species C. splendidus and C. aeruginos was collected in their sympatric area, Hwangji stream, the Nakdong river. Morphological characters as well as RAG1 gene and CO1 gene were investigated to confirm the natural hybrid origin. Both the induced hybrids and their reciprocal counterparts displayed the double peaks of sequence chromatograms at the 13 diagnostic positions. In analysis of CO1 gene, it revealed that the maternal species of the natural hybrid appeared to be C. splendidus and C. aeruginos due to their 100% sequence identity. Thus, such result gave convincing evidence for the occurrence of a natural interspecies hybrid between C. splendidus and C. aeruginos. Therefore, the C. speldndisus populations have been introduced in the Nakdong river at recent years.
