This study was done for understanding ecological and genetic perspectives of Pyrocoelia rufa (Coleoptera: Lampyridae). This study also referred field-surveying data including organic matter in soil, temperature, humidity, elevation, slope, aspect and so on, and a genetic approach using two loci on the mitochondria DNA. Totally 10 regions in South Korea were selected as target areas, and 10 individuals were sampled at each site from 2013 to 2014. Pyrocoelia rufa was investigated to prefer the region have relatively lower elevation (150m in average) and lower slope (<10 degree in average), which commonly keeps the cool temperature (under 30℃ in max) due to the dense vegetation. In aspect, there was no common characteristics among the study areas. Only what I recognized in this study was the target species had an inclination to prefer the northwestern and/or northeastern areas for its living. Cytochrome oxidase subunit Ⅱ and 16S rRNA genes which were loci of mitochondrial DNA were used for the genetic analysis in this study. The result was showing that there were three major genetic groups; two different genetic groups were divided based on a great mountain belt called Baekdudaegan (BDDG), which across South Korea from the northeastern region to the southern region; the other genetic group (in Jeju island) was divided from the mainland due to the long-term isolation. Interesting point was that Gurye county, the most southern region of BDDG, was found to commonly share the haplotypes which eastern and western sites of BDDG contained separately. This region is spatially at a border line between the east and the west of DBBG, and it can be possibly estimated that some individuals at one site move to an opposite site through Gurye county and share their DNA with each other. In conclusion, ecological and genetic approaches were needed to inspect the environmental condition for the habitat of Pyrocoelia rufa, and the possibilities and characteristics of sharing the genetic factors among several regions in South Korea. Through the field survey and genetic approach, there was recognized to be several different regions in environmental and genetic points. The results could be useful to contribute the ecological restoration, reintroduction and sustainable maintenance of the firefly.
