In this study, we designed and produced PCR Micro Fluid Chip(PCR Chip) by using polypro-pylene, O.P.P tape, double-sided tape which are easy to get because they already have mass produc-tion systems. We made the heating pattern on this PCR Chip based on PCB in order to supply thermal energy needed for biochemical experiment, and constructed Correction System to measure the heat- ing temperature precisely. Then, we verified the performance through the experiment of Quadruple PCR. At least three things have to be considered in order to produce the miniaturized PCR Chip. First, the heating method to increase the temperature of PCR Chip has to be required and the things made by this heating system have to be produced in large quantities and be made in lower cost so that it can be disposable, and the heating surface and the material have to be stuck together. It should also be able to be converted to Field Scanning System by making heating and cooling time faster using Low Energy Consuming Electric System. To meet these requirements, we used PCB(Printed Circuit Board) which has existing mass production system, thus it could be made disposable. We also made the thickness of the chip 200µm by making heating circuit with PCB. In this way, we could make the heating and cooling time shorter and get to the ideal temperature using only 5V, the power of USB. Second, reproducibility and sensitivity have to be secured to make PCR Chip commercialized. For this, we have to measure the temperature precisely and the sensor has to be corrected. Thus, although we can use various ways to measure the temperature of PCB heated by PCB Heating Circuit which was made earlier, we ultimately chose NTC Thermistor with 1% error rate instead of RTD in this study. In the fourth chapter, we explained the reason why we chose NTC Thermistor and the way we corrected the temperature. Third, the material for Chamber that contains samples including DNA or RNA is also im-portant. By this time, chips made of various materials such as silicon, glass, PDMS, PC, PET, etc. have been developed. However, it is reported that the chip made of silicon or glass has a problem because it is too expensive to be made disposable. To solve this problem, the research has been con-ducted using a high molecular substance as an alternate material. PDMS has been most commonly used as the high molecular substance in researches but we have so many difficulties in making the scale of a research bigger for commercial purposes. In this study, we used double-sided tape, O.P.P tape and polypropylene, produced in large quan-tities and widely used, as the material for Chamber instead of PDMS that has been developed for previous PCR Chip. We checked the volume of the band by conducting a PCR experiment in order to know whether 8 kinds of double-sided tapes in various materials, which we can get easily around us, are suitable for the material of Chamber. Then, we conducted quadruple PCR experiments to verify the three selected double-sided tapes. We also conducted experiments to check the temperature inside Chamber for extra researches. Because, although the sensor in NTC Thermistor will measure the temperature as the temperature is going up by the heating pattern on the surface below PCB, the measured temperature is only the temperature measured by the sensor in NTC Thermistor and we do not know whether the temperature is the same as that of DNA sample. We conducted another experiment to check fluorescence so that we can ensure the chip we made this study can be applied to Real Time PCR. The PCR course is possible in poor conditions because PCR Chip Based On PCB, proposed in this study, can supply thermal energy needed for PCR experiment by using USB power. When we compared the result of the experiment using PCR things with the one of the experiment using PCR Chip Based On PCB, we can see the results are almost same. PCR Chip Based On PCB completed the PCR course within one hour and made the result faster than commercial PCR devices which need about two hours to complete the course. Also, PCR Chip Based On PCB seems to be able to be converted to Real-Time Quantitative PCR System, according to the result that fluorescence was found in the experiment to check detection of it.
