콜라겐 스펀지 (COL-S)는 생분해성, 생체 적합성 및 유연성이 뛰어나 뼈의 재생을 위한 지지체로서 이용되고 있다. 본 연구에서는 피브리노겐 (FNG) 가교화 반응을 이용하여 COL-S 지지체에 피브린 네트워크 형성하고 그에 따른 MG-63 세포의 증식, 분화 및 부착에 대한 영향을 평가하였다. 다양한 농도의 FNG에 따른 COL-S에의 피브린 네트워크 형성에 따라 지지체의 형태, 다공성, 물 흡수도 등의 변화를 살펴보았다. 전자 주사 현미경(SEM)으로 관찰한 결과 모든 농도의 피브리노겐 사용군에서 피브린 네트워크가 형성됨이 관찰되었다. 그러나 FNG 농도가 증가함에 따라 피브린 네트워크의 형성률이 증가하였지만, 고농도의 FNG에서는 다공성과 물 흡수도는 약간 감소하였다. MTS 분석, DNA 농도 및 세포의 독성확인, SEM 이미지 분석법으로 관찰한 결과 MG-63 세포가 COL-S와 COL-S/FNG 지지체 표면 등에 잘 부착되어 있음이 관찰되었다. 또한 FNG 농도를 10, 40, 80 mg/ml로 조절한 지지체에서 배양한 세포는 염기성인산화효소 (ALP) 활성이 유의성 있게 증가함을 보였다. 이러한 결과는 피브린을 이용하여 COL-S 에 네트워크를 형성하는 것이 COL-S의 뼈 재생을 위한 생체 적합성을 증가 할 수 있음을 나타낸다.

Collagen sponges (COL-S) are used as scaffolds to support osteoblasts and stimulate bone repair, because of their flexibility, biocompatibility, and biodegradability. In this study, we added fibrin networks to COL-S scaffolds via a fibrinogen (FNG) cross-linking reaction and evaluated the effects on proliferation, differentiation, and adhesion of MG-63 cells. The fibrin network formed in COL-S with various concentrations of FNG was characterized with regard to morphology, porosity, and water uptake ability. Successful fibrin network formation was observed by scanning electron microscopy (SEM). As the FNG concentration increased, network formation increased, but porosity and water uptake ability were slightly reduced at high FNG concentrations. The MTS assay, DNA content assay, live/dead fluorescence assay, and SEM imaging showed that MG-63 cells attached and spread on COL-S and COL-S/FNG scaffolds, particularly in scaffolds modified using FNG. In addition, alkaline phosphatase (ALP) activity was also significantly increased in cells cultured on scaffolds modified using 10, 40 and 80 mg/ml FNG. Thus, the addition of a fibrin network could increase the biocompatibility of COL-S for bone regeneration.
