클로렐라에는 단백질, 아미노산, 비타민, 미네랄과 같은 영양성분뿐만 아니라 엽록소, 베타카로틴, 루테인 등의 항산화 물질이 풍부함에도 불구하고, 클로렐라 관련 기능성 연구는 대부분 건강기능식품으로 연구가 진행되어와 화장품 소재로써의 연구는 매우 적은 편이다. 본 연구에서는 그 동안 건강식품으로만 사용되어온 클로렐라를 항노화 화장품 소재로써의 가치를 파악하기 위한 연구를 진행하였다. 특히 클로렐라의 성분 중 가장 함량이 높은 단백질 (60% 이상) 성분에 주목하여, 클로렐라 내 단백질을 추출하고, 이를 효소로 가수분해하여 LC-MS로 분석하고, 데이터베이스 프로그램을 사용하여 펩타이드 위주로 물질 스크리닝을 하였다. 클로렐라의 추출 효율을 향상시키기 위해 기존에 사용되어온 열수추출 방법과 달리 효소 처리 후 추출하는 방법을 실시 하였으며, 세포벽 분해 효소와 6종류의 단백질 분해효소를 사용하여 최적의 효소 처리 조건을 제시하였다. 세포벽 효소 (YL-NL)로 1차 효소반응 시킨 후 2차로 6종류의 단백질 분해효소로 각각 효소반응 시켜서 6개의 클로렐라 추출물 시료 (Protex 6L; Sample 01, Protex 7L; Sample 02, Collupulin MG; Sample 03, Alcalase 2.4L; Sample 04, Bromelain 1200GDU; Sample 05, Pepsin; Sample 06)를 제조 하였다. 이들 시료는 Melan-a 세포에서 세포독성을 보이지 않았으며, 멜라닌생성 억제능은 Sample 02에서, 활성산소종 제거능은 Sample 01, 02, 03에서 통계적으로 유의한 수준의 효능이 있음을 알 수 있었다. 시험물질이 mushroomtyrosinase 효소의 활성을 직접적으로 억제하거나 DPPH radical을 소거하는 활성은 나타나지 않았으나 Sample 02의 경우 세포 내 멜라닌 생성을 억제하였다. 또한 시험물질의 UV에 의한 HaCaT 세포의 산화적 손상을 보호하는 효과를 측정한 결과 Sample 01, 02, 03의 경우 UVB에 의해 증가된 활성산소종의 생성을 유의적으로 감소시켰다. 각각의 시료를 HPLC, LS-MS (Q-TOF)를 이용하여 분석하고, 분석한 데이터를 Agilent Mass Hunter Qualitative Analysis software를 이용하여 peptides 분석을 하였다. 데이터베이스 분석 결과 positive mode에서는 Sample 01, 03, 06에서 Asp-Gln-Cys로 추정되는 tripeptide가, Sample 02에서 Cys-Ala-Ser으로 추정되는 tripeptide를 찾았다. Chlorella vulgaris 건조분말을 세포벽 분해효소인 YL-NL로 처리한 후, 단백질 분해효소로는 Protex 7L (60°C, pH 7.0)로 처리한 Sample 02 시료가 미백, 항산화 효능 및 기능성 물질을 추정하는데 가장 좋은 시료로 판단 되었다. Sample 02로부터 분리 정제된 클로렐라 유래 펩타이드를 LC-MS/MS 및 NMR로 구조 분석한 결과 Cys-Ala-Ser (CAS) tripeptide로 확인되었고, 안전성 효능평가 실험을 실시한 결과 cell viability에 영향을 주지 않는 것을 확인하였다. CAS tripeptide의 항산화능을 평가하고자 DPPH 자유라디컬 소거능과 SOD 유사활성을 측정하였다. 그 결과 40% 이상의 DPPH 자유라디칼 소거능을 보였고 SOD 유사활성도 약 20% 이상 나타나 CAS tripeptide가 항산화 효과를 보임을 알 수 있었다. Chlorella, a unicellular green algae, contains not only nutrients such as proteins, amino acids, vitamins, and minerals, but also substances with potential anti-aging effects including chlorophyll, beta-carotene, and lutein. However, most research on chlorella has focused merely on its role as a functional food. There are a very few studies on its function as a cosmetic material. This study investigated the cosmetic value of chlorella. In particular, this study focused on a protein that accounts for the highest rate (more than 60%) of chlorella for development of a functional cosmetic material. Traditionally, chlorella has been extracted using hot water or organic solvents. To improve the extraction efficiency of hydrolysate from chlorella, this study reviewed extraction methods after enzymatic treatment and suggested optimal enzymatic treatment conditions. Six chlorella hydrolysates, which were first extracted using cell wall lytic enzyme (YL-NL) treatment and then proteolytic enzyme treatment, were used to test whitening efficacy and anti-aging efficacy in accordance with typical evaluation methods for functional cosmetic products in human skin cells. Six kinds of proteolytic enzymes, Protex 6L (Sample 01), Protex 7L (Sample 02), Collupulin MG (Sample 03), Alcalase 2.4L (Sample 04), Bromelain 1200GDU (Sample 05), and Pepsin (Sample 06), were used in this experiment. Not all samples were shown cytotoxicity up to 100 ppm in Melan-a cell. The ability to remove active oxygen species was found in Samples 01, 02, and 03, and the efficacy was statistically significant. The test materials neither inhibited the activity of mushroom tyrosinase enzyme directly nor removed 1,1-diphenyl 2-picrlhydrazyl (DPPH) radical. However, Sample 02 inhibited melanin production, and Samples 01, 02, and 03 restricted ROS generation by UV rays. With regard to Melan-a cell and measured melanin inhibition efficacy in cells, in Sample 02, with a rise in concentration, melanin content decreased significantly. In other samples, melanin inhibition was not observed. In addition, Sample 02 had a tyrosinase inhibition effect. Each sample was analyzed via HPLC and LS-MS (Q-TOF), and the results were used for the analysis of peptides with Agilent Mass Hunter Qualitative Analysis software. According to database (DB) analysis, in positive mode, a tripeptide thought to be Asp-Gln-Cys (DQC) was found in Samples 01, 03, and 06, while a tripeptide thought to represent Cys-Ala-Ser (CAS) was found in Sample 02. In conclusion, Sample 02, which was derived from proteolytic enzyme treatment of Protex 7L after Chlorella vulgaris powder was treated with YL-NL, a cell wall lytic enzyme, turned out to be the best sample in terms of whitening and anti-aging efficacy and functionality. A novel Chlorella vulgaris-derived tripeptide was purified from Sample 02 and analyzed via LC-MS/MS and NMR. The novel tripeptide was identified as Cys-Ala-Ser and had no cytotoxic effects on cell viability at 100 μM. Antioxidant capacities were determined by DPPH radical scavenging activity and superoxide dismutase (SOD)-like activity. CAS tripeptide showed over 40% DPPH free radical scavenging activity, and had around 20% SOD-like activity. CAS tripeptide was used to treat a human skin cell line and this resulted in an increase in the expression of antioxidant-related genes, such as SIRT1, Gpx1, and Catalase. The antioxidant effect of CAS tripeptide is thought to be related to the thiol group (-SH) of cysteine, one of the CAS amino acids. As another index of anti-aging activity, a wound healing assay was conducted. CAS tripeptide did not show any wound healing effect. To investigate the efficacy of CAS tripeptide on collagen production, procollagen type I C-peptide (PIP) production was measured using a PIP EIA Kit. The expression of PIP, a collagen synthesis marker, was increased by over 50% at a 50 μM concentration of CAS tripeptide. In addition, CAS tripeptide increased the expression of procollagen C-endopeptidase enhancer (PCOLCE), which is an extracellular matrix protein that enhances the activity of procollagen C-endopeptidase in a dose-dependent manner. These results suggest that CAS tripeptide might increase collagen levels in skin. These combined results suggest that CAS, a novel tripeptide derived from Chlorella vulgaris, might be a valuable biomaterial for modulating skin homeostasis due to its antioxidant properties.
