To solve these problems, the effect of DMN processing on the activity of the encapsulated drug was investigated using lysozyme (LYZ) as a model protein. Our results showed that interaction of the encapsulated drug with the biodegradable polymer, pH, temperature, and drying conditions could affect drug activity during microneedle fabrication. Further, addition of the protein stabilizer trehalose could dramatically increase the stability of the encapsulated drug.
Also, fabricated exendin-4 (Ex-4), effective for glycemic control in patients with type 2 diabetes, loaded DMN patch using FDA approved biocompatible polymer carboxymethylcellulose (CMC). For optimization of the
fabrication process factor, such as temperature, pH, polymer CMC viscosity and CMC interaction with Ex-4 was investigated. Ex-4 loaded DMN showed no cytotoxicity and storage activity of 93.2% of Ex-4 after 8 weeks at 4°C. In vivo administration of Ex-4 loaded DMN and 2 month storage Ex-4 loaded at 4°C to diabetic mice resulted in same manner as SC injection glycemic control showing based on the pharmacodynamics studies, the Ex-4 loaded DMN showed similar satisfactory drug delivery in diabetic mice. As a result, Ex-4 loaded DMN which is encapsulating Ex-4 with maximizing stability constructed a therapeutic effect could be useful drug delivery system to type 2 diabetes patients.
This result suggested that optimization of the DMN fabrication process is required to maintain the activity of the encapsulated drug in DMN and may thus offer promising therapeutic efficacy for volatile biological drugs by minimizing loss of activity.
