세포 내 특정 세포 소기관의 종양에 표적하여 이미징 및 약물전달이 동시에 가능한 나노입자는 암 치료에 큰 관심을 받고 있다. 본 연구에서는 응집-유도방출 (AIE, aggregation-induced emission)을 보이며 쿠마린, 펩타이드 그리고 트리페닐 포스페이트의 콘쥬게이션으로부터 형성된 미토콘드리아-표적 프로브인 AIE-Mito-TPP-C에 대해 보고한다. 나노입자 AIE-Mito-TPP-C는 물과 물을 포함하는 유기혼합용매에서 약 20 nm의 지름을 가지는 나노입자(AIE-Mito-TPP-C)의 형태로 제조되었다. AIE-Mito-TPP-C는 순수한 물에서 H-agggregation을 하여 약한 세기의 형광을 나타내었고, 물과 혼합된 유기용매에서는 J-aggregation을 하여 강한 세기의 형광을 나타내었다. 또한 AIE-Mito-TPP-C의 형광세기는 물, 메탄올, DMSO, 다이옥산, THF와 같은 용매의 donor number (DN)에 의존하였다. AIE-Mito-TPP-C를 DOX (doxorubicin)과 수소결합 및 π-π 스태킹을 통하여 함께 응집을 유도하여 또다른 나노입자 (AIE-Mito-TPP-C-DOX)를 제조하였다. AIE-Mito-TPP-C-DOX는 pH 5에서 DOX를 방출하였다. 이러한 결과로 인해 AIE-Mito-TPP-C-DOX가 미토콘드리아에 선택적으로 DOX를 전달하여 방출하는 것을 알 수 있었고, 이는 미토콘드리아에 표적 가능한 이미징 프로브 및 약물 전달 시스템으로서 응용할 수 있었다.

Herein, we report a mitochondria-targeting probe (AIE-Mito-TPP-C) formed by conjugating a triphenyl phosphine with a coumarin and a peptide, which can undergo aggregation-induced emission (AIE). The AIE-Mito-TPP-C formed homogeneous nanoparticles of ca. 20 nm diameter in pure water and mixed water/organic solvents. The fluorescence intensity of homogeneous nanoparticles induced by the H-aggregation (AIE-Mito-TPP-C-H) in pure water was relatively weak as compared to the homogeneous nanoparticles induced with J-aggregation (AIE-Mito-TPP-C-J) in mixed water/organic solvents. In addition, the fluorescence intensities of AIE-Mito-TPP-C were dependent on the donor number (DN) of solvents such as water, methanol, DMSO, dioxane, and THF. In particular, we fabricated co-assembled nanoparticles (AIE-Mito-TPP-C-DOX) from mixed AIE-Mito-TPP-C and doxorubicin (DOX). AIE-Mito-TPP-C-DOX nanoparticles were formed as co-assembled nanoparticles with DOX through hydrogen-bonds and π-π stacking. We found that the AIE-Mito-TPP-C-DOX nanoparticles selectively delivered DOX into mitochondria. Furthermore, DOX molecules from AIE-Mito-TPP-C-DOX were released at pH 5. The AIE-Mito-TPP-C as a mitochondria targeting probe and delivery device provides a unique strategy for potential image-guided therapy of cancer cells.
