I also examined the effect of nicotinamide (NAM) on mitochondrial ROS and ATP generation. It causes a decrease in mitochondrial content and mitochondrial superoxide when NAM entered in cells. In addition NAM activates SIRT1 to cause a mitophagy and decrease in mitochondrial content and ROS levels. In order to determine whether NAM reduces ROS through SIRT1-mediated mitophagy, the level of mitochondrial superoxide, mitochondria content, ATP among the cells treated with NAM and SIRT1 activator SRT1720. SIRT1 activation was found to increase the mitochondrial activity while NAM decreased it. It was concluded that NAM treatment increases NAD+/NADH ratio both in cytosol and mitochondria and thereby activates SIRT1 to induce mitophagy and loweres the activity of complex I and electrone transport in mitochondria which leads to reduction of mitochondrial ATP and ROS production.
