Testing for skin irritation and skin sensitization are essential aspects of the safety assessment of cosmetic ingredients. In this study, a three-dimensional human skin model was reconstructed from human-derived keratinocytes, and this was used to establish an alternative to animal testing for the evaluation of skin irritation. In addition, immune cells were co-cultured with the reconstructed human skin model in order to explore possibilities for use as a framework for testing skin sensitization. Keratinocytes were isolated and expanded from human skin tissue to reconstruct the human skin model and optimal three-dimensional tissue culture conditions were determined. The histological morphology, viability, skin barrier function, lipids profile within the stratum corneum and production of pro-inflammatory cytokines were analyzed in the reconstructed human skin model in order to determine its similarity to human skin. The histological morphology identified four differentiated cell layers the same as are present in normal human skin, and each layer expressed the expected differentiation markers. Furthermore, the reconstructed human skin model exhibited barrier function through the formation of a functional stratum corneum. It represent the possibility to be used for in vitro skin irritation and skin sensitization testing. For the skin irritation test, the reconstructed human skin model was treated with test materials, and the cell viability was measured in order to determine the degree of irritation. The amount, application time, application method, and washing method of the test materials was varied in order to establish the optimal conditions for the test. The accuracy of the skin irritation test was determined by using twenty different types of standard materials, as suggested by the OECD TG 439, the international guideline for skin irritation testing. These tests resulted in 80% sensitivity, 70% specificity, and 75% overall accuracy of skin irritation, compared with the UN GHS classification. These values were compatible with the criteria for skin irritation predictability using validated reference skin model (VRM), as suggested by performance standard data from the OECD TG 439. For skin sensitization testing using the skin equivalent, a THP-1 cell line, which is a human monocyte leukemia cell line previously used in in vitro skin sensitization tests, was used. Cells were co-cultured with the reconstructed human skin model, creating a system in which the sensitizers are absorbed to the skin, metabolized, and transported to immune cells, thus mimicking the in vivo situation. Test materials were used at concentrations of 0.1×, 0.5× and 1×, after calculating the IC50 concentration of the cells and activation levels of CD54 and CD86, surface antigens of the THP-1 cell line, were measured. In a skin sensitization test with seven different types of test materials, including sensitizers (1-Chloro-2,4-dinitrobenzene; Alpha-hexylcinnamaldehyde; Isoeugenol), irritants (Sodium lauryl sulfate; Benzalkonium chloride), and non-sensitive and non-irritants (Isopropanol; Glycerol), the co-culture system comprising the skin equivalent and the THP-1 cell line improved the false positive rate for irritants compared to that of the THP-1 cell line alone, which was unable to distinguish between irritants and sensitizers. These results suggest that the reconstructed human skin model is a promising model for assessing the safety of cosmetics for skin irritation and skin sensitization, it represents an alternative method to animal testing.
