The mosquito-borne chikungunya virus (CHIKV) has recently emerged as an important virus with a representative symptom of rheumatoid arthritis. Therefore, a safe, inexpensive-priced vaccine is needed to prevent chikungunya fever. The purpose of this study was to investigate the immunogenicity of CHIKV to a recombinant CHIKV E2 protein following inoculation in BALB/c and C57BL/6 mice. ELISA tests were conducted with the sera of mice inoculated with the recombinant CHIKV E2 protein and the presence of antibodies was verified. Antibodies to IgG1 and IgG2a were also formed. A comparison of the antibodies revealed a dose-dependent increase as the amount and number of inoculations increased (p < 0.05). A comparison by isotype showed a significantly higher O.D. of antibodies to IgG1 in mice inoculated with 40 μg CHIKV E2 than those of with 20 μg CHIKV E2 (p < 0.05), and a significantly higher number with 60 μg CHIKV E2 than with 20 μg CHIKV E2 (p < 0.05). However, the difference between 60 μg and 40 μg CHIKV E2 was not significant. The differences for IgG2a were similar to those for IgG1 (statistically significant differences for 40 μg vs. 20 μg CHIKV E2 and for 60 μg vs. 20 μg CHIKV E2), but significant differences were also seen for 60 μg vs. 40 μg CHIKV E2 (p < 0.05). An ELISpot test conducted using the splenocytes of mice also confirmed a significant increase in the number of immune cells that paralleled the increased number of antibodies (p < 0.05). These findings indicated that the recombinant CHIKV E2 protein correlated with immunity. Therefore, the recombinant CHIKV E2 protein shows promise as a safe and effective vaccine.
