Since the industrial revolution, mankind has built its industry and economy on the basis of fossil fuel energy. However, due to arise of problems such as limited supply, limited access, pollution, emission of greenhouse gas etc. Therefore, sustainable development based on fossil fuel has no future. To overcome these problems, many researches (e.g increasing energy efficiency, development of alternative energy etc.) are being accomplished, but fundamentally, substitute energy industry is required in order to replace the current industry. At present time, biorefinery holds substantial potential and regarded as the best to lead the future chemical industry. Biorefinery is similar to the petrochemical industry in the sense of refining crude oil to produce variety of products, but refining biomass instead of crude oil to produce biofuel and biochemical products. Existing biorefineries were able to produce chemical products based on thermo-chemical reaction process but the advantages also lay in the utilization of bioresources to produce bioproducts through bioconversion process such as saccharification and fermentation. Moreover, bioprocesses have various advantages compared to chemical process such as lower energy input, lower pollutants output, and higher reaction selectivity. In this study, biomass, the feedstock of bioconversion process, were classified based on its origin, and the future potential biomass were investigated by composition analysis. Subsequently, pretreatment process, focused on chemical pretreatment, to improve sugar conversion was also investigated. Some pretreatment studies have been excessively focused on improvement of enzymatic digestibility which led to severe reaction conditions. These have negative effects on the bioconversion process such as occurrence of inhibitory compounds as well as over degradation of target materials (cellulose or hemicellulose in biomass). Therefore, in order to induce the reaction condition, to effectively reduce the losses of potential sugars portion during the chemical pretreatment, the statistical optimization by response surface method was applied using newly designed regression model. The effects of process variable on solid loss, enzymatic digestibility and sugars conversion were investigated, and then the process optimization was performed based on the results. In addition, spent coffee grounds, high potential organic biomass, were applied in the bioconversion process, and effects of alkali type on the solid loss and sugars conversion were also investigated and optimized. In development of saccharification process, the essential biological catalyst (enzymes) was applied. To secure the effective hydrolytic enzymes (cellulase, xylanase and mannanase), screening of microbial resources was performed. Production, purification, identification and biochemical characterization were carried out using selected microbial strain, and moreover applied to biomass saccharification. The produced enzymes were compared with commercial cellulase and manannase under the same condition. As a result, the sugars conversion was higher or similar effect using the biomass which demonstrated the commercialization feasibility of microbial resources. Therefore, the biorefinery with highly active biocatalyst should have advantageous position in the production of bioproducts based on the increased efficiency. In addition, fermentable sugars were produced by enzymatic hydrolysis of pretreated biomass such as Miscanthus straw, canola straw and spent coffee grounds. The hydrolysates were applied in fermentation (as a carbon source) to produce biochemical compounds such as bioethanol, cephalosporin C and cordycepin. As a result, fermentations using the hydrolysates have similar conversion when compared with standard substance (control group), thus inhibitory effects were not found during the fermentation. Finally, the overall processes were evaluated based on mass balance. The process design and evaluation for sugars, bioethanol, cephalosporin C and cordycepin production was carried out by using SuperPro Designer. Moreover, economic feasibility of designed process was evaluated. As a result, the process for cephalosporin C production shows the highest gross margin as 73.98%, and the ROI and payback time of cordycepin were 61.72% and 1.62 years, respectively. In this study, development of biomass conversion processes, has displayed feasibility of replacing future chemical industry, and moreover, sustainable development could be available when economic feasibility is resolved in the industrial scale.
