Cortical thinning patterns in Alzheimer’s disease (AD) have been studied through conventional regional analysis. However, the coordinated variance of cortical thickness in different brain regions is investigated both at the individual and group network levels recently. In this thesis, we sought to analyze network architectural characteristics of a structural covariance network (SCN) in AD, and to show the disorganized structural covariance connectivity across the brain regions in AD. We constructed individual SCNs directly from T1-weighted MR images with structural covariance which is defined using cortical thickness similarity within different brain regions. The structural covariance connectivity showed smaller small-world value in individual network study and more randomness of group network study in AD compare to NC. Furthermore, network-based structural atrophy in mild cognitive impairment (MCI) was investigated to understand pathological course using cortical thickness. Co-atrophy network was constructed by atrophy level. Epicenters and gateways, where we defined as the diseases starts and progresses in early stage, extracted from the co-atrophy networks. The results from epicenters and gateways support the transneuronal spread model which is one of the neurodegenerative models. This thesis may provide surface-based measures to understand interaction between two regions with co-atrophy of the cerebral cortex in AD.
