Inonotus obliquus (IO) has been used as a folk medicine in Russia, Asia and North America and several biological activities such as antioxidant, anti-inflammatory, anti-viral and anti-tumour effect have been elucidated by recent studies. Thus, the objectives of this study is to investigate antioxidant and anti-inflammatory activity of IO extract and fractions and to investigate the anti-inflammatory effect of IO compounds (lanosterol, inotodiol, 3-β-hydroxylanosta-8,24-dien-21-al) in LPS-stimulated RAW 264.7 macrophage. Ethylacetate fraction showed the potent peroxyl-radical scavenging activity and reducing capacity and dichloromethane fraction displayed the strongest cellular antioxidant capacity. In addition, the most potential anti-inflammatory activity in the suppression of NO, PGE2, and cytokine production was observed in dichloromethane fraction. Among three compounds as lanosterol, inotodiol, and 3-β-hydroxylanosta-8,24-dien-21-al isolated from dichloromethane fraction, 3-β-hydroxylanosta-8,24-dien-21-al showed the strongest cellular antioxidant capacity and the most inhibitory activity on the production of NO, PGE2, and cytokines such as TNF-α, IL-1β, IL-6). Additionally, 3-β-hydroxylanosta-8,24-dien-21-al dose-dependently iNOS and COX2 expression. Furthermore, 3-β-hydroxylanosta-8,24-dien-21-al attenuated the activation of IKK and IκB activation, and the translocation of NF-κB to the nucleus in a dose dependent manner. In consequence, the potent anti-inflammatory activity of 3-β-hydroxylanosta-8,24-dien-21-al on LPS-stimulated inflammation may be due to the attenuated expression of inflammatory proteins through the down-regulation of IKK/IκB/NF-κB signaling pathway. IO extract or its dichloromethane fraction can be utilized as a noble nutraceutical agent for the development of functional food aiming at anti-inflammatory effect.
