본 연구는 대사체 통합 분석을 통해 녹차 섭취에 따른 피부광노화 조절 관련 효능 및 기작을 피부, 혈액, 간, 대장 내 내인성/외인성 대사체 및 장내미생물 변화 연구를 통해 수행하였다. 녹차는 자외선으로부터 피부를 보호 한다고 잘 알려져 있으나, 손상 회복 메커니즘은 정확하게 규명되어 있지 않은 실정이다. 따라서, 본 연구에서는 통합 대사체 분석 적 접근을 통해 장기 간 녹차 섭취와 단기 간 녹차 섭취에 따른 장내 미생물 변화와 대장, 간, 혈액, 피부 내 내인성/외인성 대사체 변화 간의 상관관계를 규명하였다. 장기 간 녹차 섭취는 자외선 조사에 의해 유발된 피부 주름, 표피 두께 증가, 콜라겐 섬유 파괴를 완화시켰다. 또한, 자외선에 의해 손상된 피부 내 유전체와 대사체는 녹차 섭취에 따라 회복이 되었으며, 이는 아스코르빅산 회로와 요소 회로와 관련이 있는 것을 확인하였다. 대장과 간에서 여러 유도체 형태의 녹차 카테킨이 검출되었고, 외인성대사체인 고리 분열 물질과 페놀산류가 검출되었다. 자외선 조사에 의하여 장내 바실리 세균이 크게 변화 하였고, 리소포스포리피드의 함량이 감소하였다. 그러나, 녹차 섭취군에서 장내 미생물 군집의 다양성이 크게 증가함을 확인하였고, 특히 알로바쿨롬, 파비박터, 루미노코카시에, 라크노스피라시에가 유의적으로 변화하였다. 이에 더하여, 녹차 섭취는 장내 내인성 대사체 변화를 크게 야기 함을 확인하였다. 이러한 내인성 대사체의 변화는 녹차 섭취에 의하여 증가한 세균의 군집과 높은 상관관계를 보였다. 장기 간의 녹차 섭취와 비슷하게, 단기간의 (7일) 녹차 섭취는 자외선 조사에 의해 유발된 홍반, 내인성 대사체 변화, 장내 미생물 군집 변화를 예방하는 데에 효과적임을 확인 하였다. 녹차의 성분 중 에피갈로카페킨 갈레이트, 카페인, 테아닌의 혼합물을 단기간 섭취 하였을 때 녹차 섭취와 비슷한 예방 효과를 나타내었다. 따라서 이 연구는 장기 간의 녹차 섭취와 단기 간의 녹차 섭취는 모두 자외선으로부터 유발된 피부광노화에 효과가 있음을 확인하였고, 이러한 효능에 장내미생물과 내인성/외인성 대사체의 변화가 관여함을 확인하였다.

While green tea is known to protect skin from ultraviolet (UV) light, underlying damage-repair mechanisms and effects on other than skin such as intestinal environment remain unclear. Thus, a multi-omics (metabolomics, metagenomics, transcriptomics, and in silico modelling) approach was used to investigate interactions among effects of diet supplementation with green tea (GTD), intestinal microbiome, and exo/endogenous metabolome changes in various bio-samples (skin, large intestine, serum and liver) of photo-aging mice in long-term (10 weeks) and short-term model (1 week). In skin, long-term supplementation of GT significantly alleviate UVB-induced skin wrinkles, epidermal thickening, and collagen-fiber destruction. Along with this, key metabolic pathways, including ascorbate metabolism and the urea cycle were the major pathways exhibiting the attenuating effects of GTD on UVB-irradiated skin. In other than skin such as large intestine and liver, various conjugates of green tea catechins such as sulfated, glucuronided, and methylated and their exogenous metabolites including ring fission products and phenolic acids were detected. When UVB exsposed, intestinal microbial classes Bacilli was discriminant and levels of lysophospholipids were decreased. However, in GTD mice we observed a significant increase in the diversity of intestinal microbiome, and enrichment in Allobaculum, Parvibacter, Ruminococcaceae, and Lachnospiraceae. Dramatic changes in many intestinal endogenous metabolites were also occurred by GTD, which showed high correlations with the bacteria enriched by GTD. Similar to long-term GT supplementation, only 7 days (short-term) of GTD significantly prevent not only UVB-induced skin erythema and endogenous metabolites changes, but also intestinal microbome (especially Bifidobacteria) changes. Similar preventing effects on changes of intestinal microbiome and endogenous metabolome were observed with supplementation of mixture compounds of GT components (epigallocatechin-gallate, caffeine, and theanine), meaning synergistic effects of GT components. From these studies, these overall studies revealed that the anti-photo-aging effects by the both long-term and short-term GT supplementation is accompanied with the changes in the intestinal microbiome and endogenous metabolome in mice.
