미생물 셀룰로오스는 식물성 셀룰로오스에 비해 우수한 순도와 기계적 특성 때문에 큰 관심을 받아왔다. 그러나 미생물 셀룰로오스는 항균성의 부재, microporosity 및 낮은 생체적합성으로 인해 생물 의학 분야에서의 잠재적인 응용에 대한 한계점이 존재한다. 이러한 한계점은 나노 물질 및 생체 고분자를 이용한 미생물 셀룰로오스 복합체를 합성함으로써 해결할 수 있다. 본 연구는 나노 물질과 생체 고분자를 이용하여 미생물 셀룰로오스 복합체를 합성하고, 궁극적으로 조직 재생 분야로의 응용을 위해 미생물 셀룰로오스에 미세 기공을 만들어 우수한 세포 침투성을 부여하기 위한 것이다. 이산화 티타늄 나노입자(TiO2Nanoparticles, TiO2 NPs)가 함유된 재생 미생물 셀룰로오스 나노 복합체는 BC의 항균력 및 조직 재생 특성을 강화하기 위한 목적으로 제조되었다. 미생물 셀룰로오스 분말을 N-methylmorpholine-N-oxide/water (NMMO/H2O) 용매에 용해시키고 TiO¬2¬ 나노 입자를 미생물 셀룰로오스 용액에 혼합한 뒤 이를 유리판 위에 붓고 applicator를 이용하여 casting 함으로써 재생 미생물 셀룰로오스-TiO2 나노 복합체 필름을 제조하였다. 복합 재료의 구조적 특징은 Transmission electron microscopy (TEM), X-ray diffraction (XRD), Energy-dispersive X-ray spectroscopy (EDS) 및 Field-emission scanning electron microscopy (FE-SEM) 분석을 통해 확인되었다. 대장균(E. coli)에 대한 항균성 연구는 재생 미생물 셀룰로오스-TiO2 나노 복합체가 포함된 성장 배지에서 유의한 감소를 보였다. 항균 메커니즘은 DCFH-DA 염색법, 글루타티온 환원 분석법 및 항균 처리된 미생물의 형태학적 연구를 통해 조사되었다. 그 결과, 항균력은 복합체에 존재하는 TiO¬2 나노 입자에 의해 생성된 반응성 산소 종과 TiO¬2 나노 입자에 의한 세포막의 손상 때문인 것으로 밝혀졌다. 재생 미생물 셀룰로오스 TiO2 나노 복합체는 항균성 외에도 동물 섬유아세포 (fibroblast cell)에 대해 독성 영향이 없고 세포 부착 및 증식 능력이 뛰어났으며 복합체의 표면은 배양 7일 이내에 완전히 덮였다. 이러한 재생 미생물 셀룰로오스-TiO¬2 나노 복합체는 우수한 살균성, 생체적합성을 보여주었으며 이는 의료 분야, 특히 상처 치료 및 조직 재생 분야에 효과적으로 적용될 수 있다. 재생 미생물 셀룰로오스-TiO2 나노 복합체의 우수한 생물학적 특성을 이용하여 체외(in vitro)에서의 조직 재생 응용을 위해 이용하기 위해 재생 미생물 셀룰로오스 지지체를 합성하였다. 이를 위해, 미생물 셀룰로오스를 NMMO/H2O 용매에 용해시키고 기공 생성물질(porogen)인 염(salt)을 첨가한 후 틀(mould)에 부어 물에 담갔다. 합성된 지지체의 특성은 Fourier-transform infrared (FT-IR) spectroscopy 및 FE-SEM을 이용하여 분석하였다. Water-in-oil emulsion 방법을 이용하여 제조된 둥근 형태의 gelatin microspheres를 porogen으로 이용하였고, 이는 casting된 재생 미생물 셀룰로오스 지지체를 고온의 물에 담그면 지지체로부터 porogen인 Gelatin microsphere가 용해되어 용해된 젤라틴에 의해 지지체의 표면이 개질되는 원리를 이용하였다. 지지체의 특성은 FE-SEM, FT-IR 및 Elemental analyzer(EA)를 통해 분석되었다. FE-SEM 분석을 통해 균일한 미세 다공성을 확인하였고, FT-IR 및 EA 분석을 통해 porogen으로 이용된 젤라틴에 의해 셀를로오스가 성공적으로 개질되었다는 것을 확인하였다. 또한 지지체의 높은 porosity 및 swelling ratio 그리고 느린 water release rate는 조직 재생 응용 분야에서의 적합성을 보여주었다. Bacterial cellulose (BC) has received tremendous interest owing to its purity and better mechanical properties compared to plant cellulose. However, its potential applications in the biomedical fields are threatened by its lack of antimicrobial properties, microporosity and limited biocompatibility. These limitations are addressed through the synthesis of BC composites with nanomaterials and biopolymers. The present study was aimed to synthesize BC composites with nanomaterials and biopolymers and also impart microporosity to the BC films for better cell infiltration that will ultimately lead to tissue regeneration applications. Regenerated bacterial cellulose (rBC) nanocomposites with titanium dioxide nanoparticles (TiO2 NPs) were prepared with the aim to enhance the bactericidal activity and tissue regeneration properties of BC. Powdered BC was dissolved in an NMMO/H2O system, TiO2 NPs were mixed into the BC solution followed by casting and solvent removal. The nanocomposites were characterized through TEM, XRD and Energy-dispersive X-ray spectroscopy (EDS) assisted field-emission scanning electron microscopy (FE-SEM) analysis. Antibacterial studies against E. coli revealed a significant reduction in bacterial growth while the mechanism of action against bacteria was detected through DCFH-DA staining assay, glutathione reduction assay, and morphological study of treated bacterial cells. The rBC-TiO2 nanocomposites also showed impressive cell adhesion and proliferation capabilities with animal fibroblast cells. These rBC-TiO2 nanocomposites demonstrate potential bactericidal and biocompatible properties and could be effectively applied in the medical field, specifically for wound healing and tissue regeneration applications. The impressive biological properties of the rBC-TiO2 nanocomposites encouraged us to apply our rBC films for tissue regeneration applications in vitro. Therefore, we sought to synthesize rBC scaffolds for application in tissue regeneration. For this purpose, BC was dissolved in NMMO, and salt crystals were added as porogens, followed by casting in molds and incubation in water. The synthesized scaffolds were characterized using FT-IR spectroscopy and FE-SEM. The FT-IR spectra exhibited bands characteristic for BC in rBC scaffolds, indicating no alteration in chemical structure, while FE-SEM revealed a porous structure of the rBC scaffold. The scaffolds exhibited very high swelling ratio, indicating enhanced water absorption and nutrient exchange capacity. The in vitro biocompatibility of the rBC scaffolds was tested based on the adhesion, growth, and proliferation of animal fibroblasts (NIH 3T3), and the osteogenesis of animal osteoblasts (MC3T3-E1). Results indicated good cell adhesion, penetration, and proliferation. Alkaline phosphatase (ALP) activity and Alizarin red staining (ARS) revealed osteogenic differentiation of animal osteoblasts on the scaffolds. To further enhance the biocompatibility of rBC scaffolds, we synthesized 3D microporous regenerated bacterial cellulose-gelatin (3D rBC-G) composite scaffolds through the same casting and leaching approach. FE-SEM images showed the presence and interconnectivity of pores, while FT-IR and XPS spectra confirmed the composite chemistry of the scaffolds. In vitro biological tests showed that animal fibroblast cells (NIH 3T3) adhered to and proliferated well on the rBC-G composite scaffolds. Cell penetration assessed by Confocal microscopy indicated up to 200 µm infiltration after 7 days of incubation, suggesting the suitability of the scaffolds for three-dimensional cell culture. The enhanced expression of metalloproteases (MMPs) showed that prolonged cell incubation can lead to extracellular matrix (ECM) production inside the 3D rBC-G scaffolds. These results demonstrated that our 3D rBC-G composite scaffolds are candidates for future biomedical applications, including tissue regeneration. It was desirable to extend our rBC scaffold research to in vivo tissue regeneration applications. Therefore, we extended our study to the fabrication of porogen induced surface modified 3D microporous regenerated bacterial cellulose/ gelatin (3DMP rBC/G) scaffolds for skin regeneration applications in vivo. Circular shaped gelatin microspheres (GMS) were utilized as porogen. FE-SEM analysis confirmed the regular microporosity while FT-IR and elemental analysis confirmed the successful surface modification of cellulose with gelatin. In vitro tests showed good adhesion and proliferation of Human Keratinocytes (HaCaT) on the 3DMP rBC/G scaffolds during 7 days of incubation. Confocal microscopy showed the penetration of HaCaT cells into the scaffolds up to 300 µm depth. In vivo skin regeneration experiments in experimental mice showed complete skin regeneration within two weeks. These results indicate that our 3DMP rBC/G scaffolds are future candidate materials for skin regeneration applications in vivo. It was concluded that our technique can not only be applied to fabricate BC composites with nanomaterials and biopolymers but also 3D BC scaffolds receiving tremendous applications in biomedical fields. Following the method, 3D BC scaffolds with controlled microporosity, biocompatibility and size and shape could be fabricated for various biomedical applications specially tissue regeneration.
Bacterial cellulose (BC) has received tremendous interest owing to its purity and better mechanical properties compared to plant cellulose. However, its potential applications in the biomedical fields are threatened by its lack of antimicrobial properties, microporosity and limited biocompatibility. These limitations are addressed through the synthesis of BC composites with nanomaterials and biopolymers. The present study was aimed to synthesize BC composites with nanomaterials and biopolymers and also impart microporosity to the BC films for better cell infiltration that will ultimately lead to tissue regeneration applications. Regenerated bacterial cellulose (rBC) nanocomposites with titanium dioxide nanoparticles (TiO2 NPs) were prepared with the aim to enhance the bactericidal activity and tissue regeneration properties of BC. Powdered BC was dissolved in an NMMO/H2O system, TiO2 NPs were mixed into the BC solution followed by casting and solvent removal. The nanocomposites were characterized through TEM, XRD and Energy-dispersive X-ray spectroscopy (EDS) assisted field-emission scanning electron microscopy (FE-SEM) analysis. Antibacterial studies against E. coli revealed a significant reduction in bacterial growth while the mechanism of action against bacteria was detected through DCFH-DA staining assay, glutathione reduction assay, and morphological study of treated bacterial cells. The rBC-TiO2 nanocomposites also showed impressive cell adhesion and proliferation capabilities with animal fibroblast cells. These rBC-TiO2 nanocomposites demonstrate potential bactericidal and biocompatible properties and could be effectively applied in the medical field, specifically for wound healing and tissue regeneration applications. The impressive biological properties of the rBC-TiO2 nanocomposites encouraged us to apply our rBC films for tissue regeneration applications in vitro. Therefore, we sought to synthesize rBC scaffolds for application in tissue regeneration. For this purpose, BC was dissolved in NMMO, and salt crystals were added as porogens, followed by casting in molds and incubation in water. The synthesized scaffolds were characterized using FT-IR spectroscopy and FE-SEM. The FT-IR spectra exhibited bands characteristic for BC in rBC scaffolds, indicating no alteration in chemical structure, while FE-SEM revealed a porous structure of the rBC scaffold. The scaffolds exhibited very high swelling ratio, indicating enhanced water absorption and nutrient exchange capacity. The in vitro biocompatibility of the rBC scaffolds was tested based on the adhesion, growth, and proliferation of animal fibroblasts (NIH 3T3), and the osteogenesis of animal osteoblasts (MC3T3-E1). Results indicated good cell adhesion, penetration, and proliferation. Alkaline phosphatase (ALP) activity and Alizarin red staining (ARS) revealed osteogenic differentiation of animal osteoblasts on the scaffolds. To further enhance the biocompatibility of rBC scaffolds, we synthesized 3D microporous regenerated bacterial cellulose-gelatin (3D rBC-G) composite scaffolds through the same casting and leaching approach. FE-SEM images showed the presence and interconnectivity of pores, while FT-IR and XPS spectra confirmed the composite chemistry of the scaffolds. In vitro biological tests showed that animal fibroblast cells (NIH 3T3) adhered to and proliferated well on the rBC-G composite scaffolds. Cell penetration assessed by Confocal microscopy indicated up to 200 µm infiltration after 7 days of incubation, suggesting the suitability of the scaffolds for three-dimensional cell culture. The enhanced expression of metalloproteases (MMPs) showed that prolonged cell incubation can lead to extracellular matrix (ECM) production inside the 3D rBC-G scaffolds. These results demonstrated that our 3D rBC-G composite scaffolds are candidates for future biomedical applications, including tissue regeneration. It was desirable to extend our rBC scaffold research to in vivo tissue regeneration applications. Therefore, we extended our study to the fabrication of porogen induced surface modified 3D microporous regenerated bacterial cellulose/ gelatin (3DMP rBC/G) scaffolds for skin regeneration applications in vivo. Circular shaped gelatin microspheres (GMS) were utilized as porogen. FE-SEM analysis confirmed the regular microporosity while FT-IR and elemental analysis confirmed the successful surface modification of cellulose with gelatin. In vitro tests showed good adhesion and proliferation of Human Keratinocytes (HaCaT) on the 3DMP rBC/G scaffolds during 7 days of incubation. Confocal microscopy showed the penetration of HaCaT cells into the scaffolds up to 300 µm depth. In vivo skin regeneration experiments in experimental mice showed complete skin regeneration within two weeks. These results indicate that our 3DMP rBC/G scaffolds are future candidate materials for skin regeneration applications in vivo. It was concluded that our technique can not only be applied to fabricate BC composites with nanomaterials and biopolymers but also 3D BC scaffolds receiving tremendous applications in biomedical fields. Following the method, 3D BC scaffolds with controlled microporosity, biocompatibility and size and shape could be fabricated for various biomedical applications specially tissue regeneration.
