백신 및 항체는 인위적으로 획득된 면역계에서 중요한 역할을 한다. 그러나 백신 접종과 수동 항체 치료에 의해 제공되는 그들의 보호는 반복적인 치료 또는 고용량을 필요로 하며, 치료 비용을 증가시킨다. 형질전환 식물은 치료의 이러한 단점에 대한 경제적인 대안이다. 바이러스 성 출혈성 패혈증 바이러스 (VHSV)와 돼지 유행성 설사 바이러스 (PEDV)는 돼지와 어류 양식 산업에서 심각한 경제적 손실을 초래한다. 현재 유럽 및 아시아에서 여러 가지 허가 된 PEDV 백신을 사용할 수 있지만 VHSV 치료를 위한 상용 백신이나 치료법은 없지만 필요로 한만큼 효과적인 것으로 입증되지 않았다. 여기에서 우리는 VHSV 및 PEDV의 에피토프 영역만을 포함하거나 콜레라 독소 B 서브 유니트 (CTB)와 융합 된 부분 당 단백질 유전자의 발현을 기초로 재조합 VHSV 및 PEDV 백신 후보 물질을 개발하고, 또한 쥐 IgM- 유사 중합체IgG (mPIGS) 을 다양한 식물 숙주에서 생산할 수 있다. 쥐의 경구 면역화, 식물 유래 CTB-S1D 융합 단백질은 S1D- 특이 계통적 IgG 및 특이적sIgA 반응을 4 주째 분변에서, 6 주째에서 정점에 이르게 하였다. 또한, 정제된 PEDV-mPIGS 면역 복합체로 피하 면역시킨 마우스는 또한 중요한 항원 특이적인 혈청 IgG 항체 반응 및 세포 면역 반응을 유도하였다. 이러한 결과는 식물유래의 재조합 CTB-S1D 융합 단백질과 PEDV-mPIGS 면역 복합체가 PEDV 감염을 예방하기 위한 백신 후보 물질로 사용될 수 있음을 시사한다. 예방 접종 외에도 항원 특이 IgG 항체를 이용한 수동 면역은 감염으로부터 보호할 수 있는 또 다른 방법이다. 본 연구에서는 인간의 치주 병의 주요 원인 인 P. gingivalis에 대한 벼 유래 단일 클론 항체 (mAb)를 생산했다. 이들 단일 클론 항체는 벼 세포 현탁 배양을 이용하여 마이너 섬모 mfa1 특이적 단일 클론 항체의 발현을 기초로 하여 생산되었다. P. gingivalis A7A1-28 균주의 mfa1 단백질 53kDa에 대한 단일 클론 항체를 암호화하는 상보적 DNA를 역전사중합 효소 연쇄 반응 방법을 사용하여 항체를 발현하는 하이브리도마 세포주로부터 클로닝 하였다. mfa1 특이 적 단일 클론 항체의 중쇄 및 경쇄 유전자는 벼 α- 아밀라아제 3D 프로모터의 제어하에 식물 발현 벡터에 제작되었다. 상기 재조합 식물 발현 벡터를 입자충격법 (particle bombardment transformation)을 이용하여 벼 세포 (Oryza sativa L. cv. Dongjin)에 도입하였다. 형질전환된 벼세포는 게놈 DNA PCR 분석에 의해 검출되었으며 세포 현탁 배양을 확립하는 데 사용되었다. Western blot 분석은 mfa1에 특이적인 단일 클론 항체의 중쇄 및 경쇄가 IgG의 전체 크기로 조립되어 벼 세포 현탁 배양 물에서 성공적으로 분비되었음을 보여 주었다. 또한, 벼에서 생산된 단일 클론 항체는 대장균에서 유래된 53-kDa mfa1 단백질과 P. gingivalis의 초음파 처리된 추출물과 특이적으로 반응하였다. 이러한 결과는 벼 세포 현탁 배양에서 생산 된 mfa1 특이적 단일 클론 항체가 박테리아 단백질에 대해 생물학적으로 활성을 가지며 P. gingivalis에 의해 유발된 치주병을 예방하기 위한 수동 면역에 사용될 수 있음을 시사한다.

Vaccines and antibodies play an important role in the artificially acquired immune system. However, their protection offered by vaccination and passive antibody therapy requires repeated treatment or high doses, and lead to increased costs in the therapy. Transgenic plants represent an economical alternative solution for these disadvantages of the therapy. Viral hemorrhagic septicemia virus (VHSV) and porcine epidemic diarrhea virus (PEDV) have caused significant economic losses in the pig and fish farming industries. Currently, several licensed PEDV vaccines are available in Europe and Asia, but they have not proven to be as effective as needed, while there is no commercial vaccine or therapy is available for treatment of VHSV. Here, we develop the recombinant VHSV and PEDV vaccine candidates based on the expression of the partial glycoprotein genes containing epitope regions of VHSV and PEDV alone or fused with cholera toxin B subunit (CTB) as well as conjugated with murine IgM-like polymers of IgG (mPIGS) in various plant hosts. Following oral immunization of mice, the plant-derived CTB-S1D fusion protein induced both S1D-specific IgG and sIgA antibody responses at week 4 and peaked at week 6. Additionally, mice subcutaneously immunized with purified PEDV-mPIGS immune complexes were also induced significant antigen-specific serum IgG antibody responses and cellular immune response. These results suggest the plant-derived recombinant CTB-S1D fusion protein and PEDV-mPIGS immune complexes can be used as vaccine candidates to prevent PEDV infection. Besides vaccination, the passive immunization with antigen-specific IgG antibody is another way to protect against infection. In this study, we produced rice-derived monoclonal antibodies (mAbs) against P. gingivalis, which is the leading cause of periodontal disease in human. These mAbs were produced based on the expression of the minor fimbriae mfa1-specific mAbs by using rice cell suspension culture. The complementary DNA encoding monoclonal antibodies to 53-kDa of mfa1 protein of P. gingivalis A7A1-28 strain was cloned from hybridoma cell lines expressing the antibodies using reverse transcription-polymerase chain reaction method. The heavy and light chain genes of mfa1-specific mAbs were constructed in plant expression vectors under the control of the rice α-amylase 3D promoter. The recombinant plant expression vectors were introduced into rice cells (Oryza sativa L. cv. Dongjin) using particle bombardment transformation method. Transgenic rice calli were detected by genomic DNA PCR analysis and used to establish cell suspension cultures. Western blot analysis showed the heavy and light chain of the mfa1-specific mAbs were assembled into full-size of IgG and successfully secreted into rice cell suspension culture medium. Furthermore, rice-produced mAbs showed specific reactivity with the E. coli-derived 53-kDa mfa1 protein and sonicated extracts of P. gingivalis. These results suggest that mfa1-specific mAbs produced in the rice cell suspension cultures are biologically active against the bacterial protein and can be used for passive immunization to prevent P. gingivalis-induced periodontal disease.
