Mechanistically, fingolimod is phosphorylated in vivo to produce fingolimod phosphate (fingolimod-P), which acts as a sphingosine 1-phosphate (S1P) receptor modulator with high binding affinity to four of the five known S1P receptors (S1P1, S1P3, S1P4 and S1P5). Although it shows high efficacy for treatment of MS, it has been reported that its low selectivity, in particular, due to high binding affinity to S1P3 causes unfavorable side effects such as bradycardia. Therefore, S1P1 selective agonists with high efficacy are highly demanded for development of new MS drugs. The therapeutic strategy towards S1P1 receptor agonists in this thesis is to lower the circulating lymphocytes more efficiently by internalization of S1P1 on lymphocyte and to enhance remyelination by modulation of S1P5 on oligodendrocytes. For this purpose, a series of the new S1P1 receptor agonists was designed based on the structures of ONO-4641 and BAF312, which were recently developed as selective S1P1 receptor agonists by pharmaceutical companies. The initial study in my laboratory identified KKSM07016 as a first hit compound showing good agonistic activity against the S1P1 receptor. In addition, it was confirmed that it fits well to the binding site derived from the X-ray crystal structure of the reported ligand-receptor complex. In this study, synthesis and biological evaluation of a new series of S1P1 agonists having different core structures such as tetrahydronaphthalene, indane and chromane are described. A total number of 24 new compounds were synthesized. Among them, KKSM0824 shows the highest agonistic efficacy with EC50 value of 0.157 μM in the calcium immobilization test. This compound will be further optimized as a potential candidate for development of a selective S1P1 receptor agonist.
