BACKGROUND: Glycolic acid peel is a popular superficial peeling technique associated with low morbidity and prompt recovery. These processes, as well as the final step of corneodesmolysis, which mediates exfoliation, are often disturbed by environmental conditions, resulting in dry, flaky skin conditions. Therefore, aftercare is required to avoid these side effects and achieve the maximum possible improvement in skin condition. OBJECTIVE: To evaluate the effect of epidermal growth factor and palmitoyl pentapeptide-4 after glycolic acid peeling. Palmitoyl pentapeptide-4 and rhEGF maintain skin barrier function and show excellent regeneration effects. METHODS: A base gel, a gel containing rhEGF, and a gel containing palmitoyl pentapeptide-4 were prepared using a two-step process, and their safety was evaluated by patch test. Regarding stability, the physical characteristics of the gel samples (composition stability, heat stability, viscosity, pH, discoloration, and odor) were tested. The gels were applied to the cheeks of 30 healthy female volunteers (age 25–39 years) for 4 weeks. Subsequently, the volunteers underwent a glycolic acid face peel. Glycolic acid (20%) was applied topically to the face for 10 minutes once per week for 4 weeks. The volunteers were then randomly assigned to apply the base gel, gel containing rhEGF, or gel containing palmitoyl pentapeptide-4 to their entire face twice per day, and instructed not to use other medications, including topical agents, for the 4-week application period. Stratum corneum hydration, transepidermal water loss (TEWL), and erythema index were then evaluated. RESULTS: Patch test results were interpreted according to the International Contact Dermatitis Research Group (ICDRG) guidelines. No specific skin reaction in prepared gels was detected by patch test. Gel samples were initially evaluated for physical stability using a centrifugation method. All samples remained homogeneous under these conditions, and did not change as the temperature increased. Therefore, the samples were considered to have appropriate stability. The stability of the viscosity, pH, discoloration, and odor of the gels was evaluated for 28 days.
