Transient receptor potential(TRP) 채널은 체온을 포함한 다양한 생리학적 기전을 조절하는 이온 채널로서, 이 중 일부 thermo-TRP 채널은 체외의 온도 및 여러 자연 유래 물질에 의해 활성화된다. 본 연구에서는 인삼(GR), 마황(EH), 육계(CC), 건강(ZR)의 네 가지 대표적인 온열약(溫熱藥)을 선정, 이들 본초(本草)가 thermo-TRP 채널의 조절을 통해 갈색 및 베이지 지방에서 체열 발생을 일으키는지 여부를 조사하였다. 2주간 4 종류의 본초(本草)를 처치한 후, C57BL/6 마우스의 백색 지방과 갈색 지방 조직에서 thermo-TRP 채널의 변화를 관찰한 결과, GR과 EH 처치는 TRPV1의 발현을 증가시켰으며 CC와 ZR의 처치는 TRPV1과 TRPA1 두 유전자 모두의 발현을 유의적으로 증가시켰다. 또한 온열성(溫熱性) 본초(本草)들은 갈색 및 베이지 지방의 분화 및 활성화에 중요한 역할을 하는 UCP1과 PPAR-γ coactivator 1-alpha (PGC1α), PR domain containing 16 (PRDM16)를 모두 유의하게 증가시켰다. 더불어 에너지 소비 기전의 핵심 인자인 AMP-activated protein kinase alpha(AMPK) 역시 이 본초(本草)들에 의해 증가됨을 확인하였다. 온열약(溫熱藥)이 마우스의 생리 지표에 미치는 영향을 관찰한 결과, 이 네 가지 본초(本草)는 모두 체중의 증가와 백색 지방 조직의 무게를 감소시켰으며, 갈색 지방 조직 부위의 체표 온도를 증가시켰다. 이중 CC와 ZR을 처치한 군에서는 직장 체온 역시 유의하게 상승하였음을 확인하였다. 이어 진행한 3T3-L1 백색 지방 세포와 일차 배양 갈색 지방 세포를 이용한 세포 실험에서 이 네 가지 온열약(溫熱藥)은 TRPV1과 TRPA1에 대한 조절 효과가 나타남을 확인하였으며, 발열능과 연관된 인자들 역시 유의하게 상승시키는 효과가 있었음을 확인하였다. 마지막으로 TRPV1과 TRPA1 길항제를 처치한 지방 세포에서 온열약(溫熱藥)들의 갈색 및 베이지 지방 활성화 기전이 차단됨을 확인한 결과, 본 연구에서 실험한 본초(本草)들은 thermo-TRP 채널을 경유해 갈색과 베이지 지방의 발열능 활성화를 유도함을 확인하였다. 결론적으로 본 연구는 온열약(溫熱藥)은 TRPV1과 TRPA1의 활성화를 통해 비떨림열발생 기전을 조절함을 밝혔으며, 따라서 본초(本草)의 사기(四氣)를 판단하는 표지 인자로서 thermo-TRP 채널의 활용 가능성을 제시한다.

Herbal drugs used in Traditional Korean Medicine (TKM) prescriptions possess unique characteristics, four qi (cold, hot, warmth and coolness) and five flavors (sourness, bitterness, sweetness, pungency and saltiness), which imply expected actions of the herbs. In specific, hot and warm herbs are prescribed for cool/cold diseases, as the administered patient is expected to gain heat. However, whether these herbs can actually elevate physiological temperature is still unknown. Therefore, this study attempted to provide evidence for thermogenic effect of hot/warm herbs. The brown adipose tissue (BAT) is a site of thermogenesis via action of uncoupling protein 1 (UCP1). In addition, UCP1-expressing beige adipocytes also develop in white adipose tissue (WAT) under certain stimuli. These two sites, brown and beige fat, are key regions in non-shivering thermogenesis. Transient receptor potential (TRP) channels are ion channels involved in various physiological roles, including body thermoregulation. Among TRP channels, a number of them known as thermo-TRP channels respond to stimulation by temperature or various nature-derived materials. In this study, we chose four representative herbs from the warm/hot catigorization, Ginseng Radix (GR), Ephedrae Herba (EH), Cinnamomi Cortex (CC) and Zingiberis Rhizome (ZR) and investigated whether they can activate brown/beige fat thermogenesis via thermo-TRP channels. After a 2-week-herbal administration, the thermo-TRP channel changes were evaluated in tissues of C57BL/6 mice. GR and EH administration elevated Trpv1 expression in inguinal WAT (iWAT) and BAT, while CC and ZR administration increased Trpv1 and Trpa1 in both tissues. Furthermore, we observed the changes in thermogenic-factors in mice. As a result, the herbs significantly induced the thermogenesis-related factors, UCP1 and PPAR-γ coactivator 1-alpha (PGC1α). PR domain containing 16 (PRDM16), a transcription factor of brown and beige adipocyte differentiation was elevated as well. Furthermore, AMP-activated protein kinase alpha (AMPK), a key regulator in energy expenditure was also increased by GR, EH, CC and ZR treatment. Next, the physiological symptoms were assessed. The administration of GR, EH, CC and ZR significantly reduced body weight gain and WAT weight, and even more, elevated skin temperature. In CC and ZR treated mice, core body temperature was increased as well. Further in vitro experiments using 3T3-L1 adipocytes and primary cultured brown adipocytes showed that the TRPV1 and TRPA1 regulating features of these herbs were also effective in the cellular level. Consistent to the vivo assays, treatments of the four herbs increased brown/beige adipocyte-specific thermogenic factors in both vitro models. Through experiments with TRPV1 and TRPA1 antagonists, we could confirm the thermogenesis from GR, EH, CC and ZR was via activation of these thermo-TRP channels. Overall, this study shows that warm and hot herbs induce the non-shivering thermogenic program via regulation of TRPV1 and TRPA1. In addition, our results suggest thermo-TRP channels as a possible indicator for classification of herbs.
