The purpose of this study was to research the changes of somatic embryogenesis and organ differentiation tendency according to external stress factors for Kalanchoe pinnata and to study this phenomenon histologically. K. pinnata, which is grown in the laboratory, has been known to form somatic embryos capable of asexual reproduction at the edge of leaves. The concentration of agar in the culture bottle and the plate was adjusted to 0.5% to 1.5% using stress factor. After that, the stem and leaf of K. pinnata were microtechnically processed. In order to study the development and changes of tissues in the in vitro culture, the changes of hormone and stress were tested in the same manner. In vivo, K. pinnata was found to produce 2nd and 3rd plantlets due to lack of water and excessive light conditions. The lower the concentration of agar in the culture bottle and the plate was, the faster the increase of growth individuals and biomass is. The occurrence of plantlet showed no tendency to occur, and the increase in the number of individuals showed a tendency to decrease with increasing agar concentration. Plantlet development and leaf regeneration were observed only in agar 1.5% medium. The difference of agar concentration was a stress factor in the root of K. pinnata, and the pattern of asexual reproduction tended to change accordingly. It is believed that by gene expression, it undergoes asexual reproduction in the leaf when the root receives high stress whereas it does in the stem with low stress. It is necessary that leaf regeneration in 1.5% agar should be compared with that of internal structure.
