퇴행성 뇌질환 환자의 체세포로부터 유래한 유도만능줄기세포(이하 iPSCs)는 약물 스크리닝에 유용한 모델로서 사용된다. 현재 대부분의 퇴행성 뇌질환 세포 모델은 이차원적인 환경에서 배양 및 분화된다. 그러나 이러한 이차원적 모델은 실제 인간의 뇌 내 미세환경을 반영하기에 적합한 모델이 될 수 없다. 따라서 이차원적 모델에 비해 보다 안정적으로 알려진 삼차원적인 조건으로 질환 모델을 구현하기 위하여, 마트리젤을 주입한 PDMS기반의 미세유체공학 기술을 적용하였다. 최근 연구에서, CAG염기서열 반복이 143개로 알려진 환자로부터 유도만능줄기세포를 만들었으며 이에 대한 신경학적 분석을 진행하였다. 기존의 정상적인 유도만능줄기세포 및 인간 배아줄기세포에 비하여 신경세포 분화의 초기에 불안정하고 활성이 낮은 양상을 보여 이를 보완하고자 PA6 세포와 공동 배양함으로써 신경 세포로의 분화를 진행하였다. 미세유체공학 시스템을 이용한 HD143 유래 신경세포의 분석에서, 우리는 중요한 미토콘드리아의 결함을 발견하였으며, 이로 인해 헌팅턴 병의 병리학적 특성으로 잘 알려진 미토콘드리아의 기능장애에 대하여 연구를 진행하였다. 이러한 결과에 기인하여 삼차원적인 미세 환경에서도 신경세포로 잘 분화하는 것을 확인하였으며, 실제 HD143 hiPSC에서 유도한 신경세포에 HD의 치료제로서 뉴런의 튜불린 아세틸레이션 활성을 조절하는 것으로 잘 알려진 히스톤 디아세틸레이즈 억제제(이하 HDAC6 억제제)를 처리하였다. 그 결과, HDAC6 억제제를 처리하였을 때 HD뉴런의 길이와 같은 형태학적 특성이 증진되었으며, Mito-tracker분석 및 미토콘드리아 외막에 발현하는 TOM20의 면역형광분석을 통해 미토콘드리아의 형태학적 특성 또한 향상되는 것을 확인하였다. 또한, 퇴행성 뇌질환 환자 초기 발병학적 특성으로 보고된 신경 가소 돌기의 개수 및 형태학적 변화를 관찰하였다. 이러한 결과는 미세유체공학 기술이 세포의 질병 모델의 신경학적 결함을 모니터링하고 약물 처리 이후의 영향을 확인할 수 있는 유용한 시스템으로 적용 가능함을 강하게 뒷받침 한다. 이를 토대로, 앞으로 보다 정교한 삼차원적인 시스템을 구현하고 약물 스크리닝을 위한 시냅스의 미세환경을 모사하기 위한 연구가 필요할 것이다.

At present, most cellular models for neurodegenerative diseases models are maintained under two-dimensional (2D) culture and differentiation conditions. However, this 2D model may not be a suitable model to reflect the microenvironment in real human brains. In order to realize disease models under 3D condition, which is known to be more stable and appropriate than 2D condition, we employed microfluidic technology using Matrigel-coated Poly dimethyl siloxane (PDMS). In the present study, we analyzed the neuronal properties of an hiPSC line generated from a patient of Huntington’s disease carrying 143 CAG repeats (HD143-iPSC). Although its initial neural inducing activity was lower than that of human embryonic stem cells and normal iPS cells, we differentiated HD143-iPSC into neural lineages by co-culturing with PA6 stromal cells. From our analysis on neuronal cells derived from HD143 using microfluidic system, we detected a significant mitochondrial defect. Also, we studied well known mitochondrial dysfunction among HD pathology. Based on this outcome, we confirmed the actual differentiated neuronal cell being stably differentiated under 3D condition. Based on this observation, we treated HD143 iPSCs derived neuronal cells with HDAC6 inhibitor (HDAC6i) as a possible therapeutic agent for HD, which is also known to regulate tubulin activity. As a result, we found that treatment of HDAC6i can improve the morphology of HD neurons, such as neurite lengths. We also found that mitochondria morphology was improved significantly, judged by Mitotracker analysis and Tom20 immunocytochemistry. Also the number of dendritic spines and morphological change matched with the earlier report in a neurodegenerative disease patient’s neuron. These results strongly suggest that microfluidic technology can provide useful systems to monitor neuronal defects of disease cell lines and the subsequent effect of drug treatment. Based on this result, we are going to carry out studies to build more elaborate 3D aligned system, and apply it to bundle of synapse with micro-mimic environment study for drug screening.
