헌팅턴병은 무도증, 성격변화 및 치매가 특징적인, 변성적 상염색체 우성 유전 장애이며, 염색체 4번의 단완에 있는 유전자에서 트리누클레오타이드 반복서열이 증가하여 일어난다. 현재 치료 방법으로는 명백한 치료방법은 없지만 약물을 투여하여 운동장애를 가능한 한 최소화 하며 정신증상 및 무도증을 완화시키는 것이 전부이다. 현재 손상된 신경 세포를 대체하는 방법으로 세포 치료가 많이 연구 되고 있다. 줄기 세포에서 신경세포 및 신경 교세포를 만들어 뇌에 세포 이식 치료법을 연구 개발 되고 있다. 본 연구는 난치성 신경계질환인 헌팅턴병의 동물 모델을 대상으로 세포이식을 통해 신경재생 유도 및 세포치료 가능성을 조사하고자 하였다. 사람 유도만능줄기세포 유래의 신경전구세포와 사람태아 유래의 신경전구세포를 세포치료에 이용하기 위해서 퀴놀린산을 주입하여 만든 헌팅턴병 동물 모델에 이식했으며, 이후 세포이식에 따른 동물 행동 능력 개선과 손상된 신경세포의 재생을 관찰했다. 세포이식 12주 후 이식한 세포의 효능을 분석한 결과 행동학적 호전, 병변 부위에서의 중간의 가시 투사 신경세포의 복구, 행동학적 호전과 연관된 신경세포의 형성, 내생적 신경세포들의 재생 증진, 염증반응의 감소, 손상을 입은 신경 단위의 연결의 재구성들을 일으킴을 알 수 있었다. 이와 같은 연구 결과를 통해 사람 신경계 전구세포가 헌팅턴병의 전임상 동물모델 치료에 효과적 이라는 것을 알 수 있었고 세포치료의 가능성을 확인 할 수 있었다.

Huntington’s disease is a devastating, autosomal-dominant inheritance disorder, involving the progressive loss of medium spiny neurons (MSN) and neuron function in the brain. Cell replacement therapy has been considered as a potential therapeutic strategy for human neurological disorders. In this study, I investigated the therapeutic effects of neural precursor cells derived from induced pluripotent stem cells (1231A3-NPCs) and human fetal neural progenitor cells (fetal-NPCs) in the quinolinic acid (QA) lesioned rat model of Huntington’s disease. 1231A3-NPCs and fetal-NPCs were injected into striatum at 1 week after QA lesioning and the transplanted animals were evaluated using various behavioral tests, including staircase test, rotarod test, stepping test and apomorphine-induced rotation test, for up to 12 weeks. The animals exhibited significant behavioral improvements. Transplantation of 1231A3-NPCs and fetal-NPCs partially replaced the lost neurons, enhanced endogenous neurogenesis, reduced inflammatory response, and reconstituted the damaged neuronal connections. Thus, the neural precursor cells derived from induced pluripotent stem cells and human fetal neural progenitor cells could effectively treat a preclinical animal model of Huntington’s disease.
