LPS 주사 1시간 후, 폐조직에서 PTK2의 인산화가 증가되는 것이 확인되었으며 이러한 효과는 4시간 시점에서는 소실되었다. PF-562271을 전처치 하였을 때 PTK2의 인산화는 효과적으로 억제되었다. 기관지폐포세척액의 싸이토카인은 모든 군에서 차이를 보이지 않았지만, 폐의 수분 함량과 조직학적 폐손상은 LPS 투여하였을 때 유의하게 증가하였으며, PF-562271을 투여하여도 이러한 손상은 지속되었다. 결론적으로 PTK2는 급성호흡곤란증후군의 초기에 인산화를 통하여 활성화되는 것을 확인하였고 PF-562271은 이러한 인산화를 차단하였다. 그러나 PTK2의 억제가 급성호흡곤란증후군을 완화시키는 효과는 관찰되지 않았다.

Acute respiratory distress syndrome (ARDS) is an inflammatory lung disease characterized by increased permeability pulmonary edema or alveolar flooding. Protein tyrosine kinase 2 (PTK2), also known as focal adhesion kinase (FAK) is a key molecule that regulates endothelial barrier function and vascular permeability. Previous in vitro studies indicate that inhibition of PTK2 may attenuate ARDS. C57BL/6 mice were used and ARDS was induced by intraperitoneal injection of lipopolysaccharide. PF-562271 was used as an inhibitor of PTK2 and was given orally 2 hours before LPS challenge as a premedication. Western blot analysis for PTK2 and phospho-PTK2, bronchoalveolar lavage (BAL) for cytokine analysis, wet to dry weight ratio, and histologic examinations were performed. The PTK2 phosphorylation was increased at 1 hour after LPS injection, but normalized at 4 hours. PF-562271 successfully blocked the phosphorylation of PTK2. Interleukin-6 was elevated in the BAL fluid after challenge with LPS, but PF-562271 did not attenuate inflammation of the lung. Similarly, pulmonary edema content and histologic features indicated that lung injuries occurred after LPS injection, and the injuries also occurred in the groups that received PF-562271 as a premedication. In conclusion, PTK2 activation was confirmed at early stage of ARDS, but inhibition of PTK2 did not attenuate lung injuries in the murine model of ARDS.
