Part II는 인간 콜라겐을 포함하는 기능성 창상치료제 개발 연구이다. 기존 항산화능 등 그 물성이 우수하다고 알려진 누에 유래 실크 단백질을 혼합한 지지체를 개발하고자 하였다. 먼저 누에를 동결 건조한 후 실크 단백질의 원료인 실샘(gland)만을 회수하여 수용성 실샘가수분해물(silkworm gland hydrolysate, SSGH)을 생산하였다. 인간 콜라겐과 실샘가수분해물을 적정 비율별로 혼합하고 가교제와 반응하여 3차원 지지체를 제작하였다. 제조된 3차원 지지체의 다공성(61-81%), 팽윤도(50-75%), 단백질 분해도를 측정하였다. 지지체는 세포독성이 없었으며, 활성산소 억제 효능이 우수하였다. 또한 마우스를 이용한 2주간의 창상모델 시험에서 복합지지체를 처리한 군이 대조군(Mediform)보다 빨리 치유되었다. 결론적으로, 콜라겐과 실샘가수분해물를 혼합한 복합제제는 창상치유에 매우 효과적임을 확인할 수 있었다. In the field of tissue- and bioengineering it is now well understood that applying biomaterials together with cells is a much more effective treatment than applying cells alone. Following such a tendency, advanced composite therapies compounded of cells and extracellular matrix are being developed in the field of cell treatment. The present study aims to develop a novel advanced composite therapy using collagen, the main component of the extracellular matrix, to be used in tissue engineering. To date, hyaluronic acid extracted from animal sources and collagen from pig skin, which are implantable, biocompatible, and biodegradable extracellular matrix components, have been used as single materials. However, collagen from pig skin has limitations to its use due to the induction of an immune response in 3-5% of people. In the present study, a process for extracting collagen from the placenta and umbilical cord discarded at birth is developed through disinfection, hydrolysis, and purification, resulting in the production of human collagen that evokes no immune response. This study attempts to compound human collagen with natural biomaterials in order to enhance the effects of wound treatment and tissue regeneration. "Collagen-silk peptide" composite wound treatments are developed using silk protein from silkworms, and its effects verified in animal tests (Part II). In addition, "collagen-hyaluronic acid" composite fillers are developed for the repair of subcutaneous tissue using crosslinked hyaluronic acid, and its in vivo effects verified (Part III). Part II describes research regarding the development of a functional wound treatment containing human collagen, which aims to develop scaffolds using existing silk protein that is known to possess excellent properties including antioxidant capacity. SSGH (Soluble Silkworm Gland Hydrolysate) is produced by collecting silkworm glands, the raw material of silk protein, after freeze-drying silkworms. Three-dimensional scaffolds are developed by compounding human collagen with SSGH at an appropriate ratio, and reacting with a crosslinker. The porosity (61-81%), degree of swelling (50-75%) and protein degradation of these three-dimensional scaffolds are measured. The scaffolds were found to have no cytotoxicity and showed an impressive inhibitory effect on active oxygen. In the wound-model experiment with a community of mice, which was conducted over a period of two weeks, the group treated with the composite scaffolds healed faster than the control group treated with Mediform. In conclusion, it was confirmed that the composite therapy compounded of collagen and SSGH is very effective for would treatment. Part III describes the development of a composite filler containing human collagen for the repair of subcutaneous tissue that aims to produce a filler with long-lasting effects via combination with existing crosslinked hyaluronic acid fillers. The collagen-hyaluronic acid composite fillers are produced at two different ratios of 10:1 and 5:1, and the viscosity and injection force of each filler are analyzed by comparison with each other. Subsequently, the in vivo persistence of the fillers is examined through subcutaneous injection in mice, over a period of sixteen weeks, and it was found that the composite fillers last longer. Restylene, a commercial hyaluronic acid, and the commercial pig collagen TheraFill were used in the control group. It was observed that the cells of the surrounding tissue grew into the composite fillers and new blood vessels were formed. In contrast, neither influx of cells nor the formation of new vessels were observed in the control group. Consequently, it was confirmed that the composite fillers containing human collagen developed herein are very effective in the repair of subcutaneous tissue. The present study confirms that the human collagen-containing composite biomaterial created here is superior to the existing biomaterials using minerals, and can be used as a material for wound healing. In the future, this study is expected to become a cornerstone of applicability and possibility of human collagen for the development of advanced composite therapies using biomaterials in the fields of tissue engineering and regenerative medicine.
