Polymerase chain reaction (PCR) is a molecular biology technique that can amplify the genetic information of the organism to be detected. This technique is achieved through three steps: separation, binding, and elongation of DNA with appropriate reagents and temperature control. Because the PCR results are very sensitive to temperature, accurate temperature control of the sample can ensure reliability, reproducibility and sensitivity to the results. This study was conducted to observe the temperature of the reagent in the micro PCR chip chamber of our micro PCR system using a very thin thermocouple. There was an error when comparing the measured temperature with the actual reagent temperature in the micro PCR chip, and the relationship between the two temperatures was very linear. By reducing the error of the two temperatures, the actual temperature of the reagent can be estimated by multiplying the measured temperature by the specific constant. If the ramping speed is relatively fast, the total experiment time is reduced as the time to reach the target temperature is shortened. Therefore, it is possible to measure the ramping speed of the reagent by measuring the internal temperature, so that the heating power parameter can be determined by comparing the ramping speed according to various power quantities in order to determine the optimal heating power amount. In order to determine the optimum micro PCR chip chamber thickness, we compared the ramping speed with the chamber thickness. The thinner the chamber, the faster the ramping rate was.
