Obesity has been a major public health epidemic worldwide. The severity of obesity is increasing with a corresponding increase in an importance of obesity-induced skeletal muscle loss. Kaempferia parviflora Wall. ex Baker, also called “black ginger” is known to elicit various biological activities such as anti-oxidative, anti-inflammatory, and gastroprotective effects. In this study, we investigated the effects of Kaempferia parviflora and its active compound 5,7-dimethoxyflavone (DMF) on obesity and muscle atrophy using ob/ob mice. Wild-type C57BL/6J and ob/ob mice were provided with a normal diet ad libitum. The ob/ob mice were orally administered Kaempferia parviflora extract (KPE) at a dose of 100 mg/kg/d or 200 mg/kg/d, and DMF at a dose of 20 mg/kg/d or 50 mg/kg/d for eight weeks. KPE and DMF significantly decreased body weight, fat volume, and fat weight without affecting appetite. KPE and DMF inhibited the expression of adipogenic transcription factors and lipogenic enzymes by upregulating AMP-activated protein kinase (AMPK) in epididymal fat. Also, the symptoms of obesity including hypercholesterolemia and non-alcoholic fatty liver were reduced. In contrast, KPE and DMF markedly increased the muscle fiber size, muscle volume, and muscle mass. This resulted in the enhancement of muscle function, such as exercise endurance and grip strength. On the molecular level, it activated the mammalian target of rapamycin (mTOR) pathway and inhibited forkhead box O3a (FoxO3a) by upregulating the phosphatidylinositol 3 kinase (PI3K)/Akt pathway in gastrocnemius muscle. In soleus muscle, mitochondrial biogenesis-related markers including peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), nuclear respiratory factor 1 (NRF1), mitochondrial transcription factor A (TFAM), and estrogen-related receptor alpha (ERRα) were upregulated by KPE and DMF. Taken together, these data indicate that KPE and DMF could be promising functional food materials for obesity by inhibiting adiposity and increasing muscle mass and function.
