현대의료에 있어 자기장은 자기공명영상장치와 같은 진단 기기에서부터 경두개 자기자극, 표면 연부조직의 통증과 부종에 대한 자기장 자극과 같은 치료에 적용되는 장치까지 매우 다양한 방법으로 사용되고 있다. 특히 자기장 치료는 부상을 당했을 때의 통증과 염증, 그리고 현대인의 고민 중 하나인 비만 환자에 대한 지방 제거에 대해 비침습적이고, 안전하며 쉬운 방법을 제공해주는 것으로 알려져 있다. 이러한 자기장이 인체 내에서 작용하는 메커니즘을 알아내기 위한 연구는 오랫동안 이루어져왔으며, 자기장의 생물학적 영향에 대한 가장 근본적인 질문은 전자기장 신호가 어떠한 생체물리학적인 상호작용을 통해서 세포에 인식되는 지에 대한 것이다. 세포와 조직의 기능을 조절하는 생체물리학적인 상호작용 기전 및 신호는 밝혀져야 한다. 현재까지 수행된 자기장과 관련된 연구들은 표적의 위치, 자기장의 세기, 자극 주파수, 자극시간 등이 설계한 연구에 따라 제각각이었으며, 자기장 자극 시스템에 대한 검증을 하지 않아 자기장에 의해 발현되는 효과를 정량화하는데 한계점을 가지고 있다. 본 연구에서는 자기장 신호가 생물학적으로 미치는 영향을 보다 정량적으로 접근하기 위한 자기장 발생시스템을 개발하였다. 개발된 시스템은 연골 세포 및 인간유래 중간엽 줄기세포 (hMSC)의 지방분화에 대한 최적의 자기장 자극 조건을 찾기 위해 자기장의 세기 및 자극 주파수를 조절할 수 있도록 하였으며, 세포시료 내 자기장 자극의 균일성은 컴퓨터 모의실험을 통해 예측하였다. 또한 세포 수준의 연구에 적합한 장치인지 검증하기 위해 연골세포와 줄기세포의 지방분화에 적용하여 생물학적 변화를 관찰하고 동시에 적절한 세포자극 프로토콜을 도출하고자 하였다. 세포시료 내에 균일한 자기장을 발생시키기 위해 헬름홀츠와 솔레노이드 형태가 결합된 코일을 개발하였으며, 100 Hz 이하의 시변 자기장을 만들기 위한 자기장 컨트롤 모듈, 0.3 T - 1.0 T의 범위에서 자기장 출력이 가능한 대전류회로를 제작하였다. 제작한 세포 자기장 자극 시스템으로 연골줄기세포 증식 및 인간유래줄기세포의 지방분화 적용하여 그 영향을 살펴보았다. 연골줄기세포 증식에 유효한 자극 프로토콜은 0.3 T / 10 Hz / 60분 이었고, 인간유래 중간엽줄기세포 (hMSCs)의 실험에서 지방분화 억제에 유효한 시변 자기장 자극 프로토콜은 0.3 T / 10 Hz / 10분의 자극에서 자기장 자극이 없는 그룹과 비교하여 유의한 차이를 나타내었다. 본 연구에서 제시한 자기장 자극 프로토콜은 연골세포 증식과 hMSCs의 지방분화에 각각 다른 프로토콜에서 유효한 결과를 나타내었다. 이는 자기장 자극의 대상에 따라 다른 자기장 자극이 필요함을 시사하고 있다. 그러므로 자기장 자극을 적용하여 인체나 세포의 변화를 분석하기 위해서는 본 연구에서 제안한 자극 매개변수의 변경이 가능한 자기장 자극 시스템과 같은 장치가 필요하다는 결론을 내릴 수 있었다.

In a recent medical field, various devices and related techniques using magnetic field have been used from magnetic resonance imaging system for diagnosis to transcranial magnetic stimulation (TMS) and stimulation for the pain and edema treatment of the soft-tissue surface. Particularly, it is well known that magnetic field therapy provides invasive, safe and easy treatment way for injury, pain, inflammation and removing lipid tissues of the obesity patients. Many researchers have long been conducted to discover the mechanism of the magnetic field effect in the human body, and a basic question on the biological effects of the magnetic field is that through which biophysical interaction, cell recognizes the magnetic field signal. These biophysical interaction mechanisms and signals which have abilities to control functions of cell and tissue are the subjects to be investigated. Magnetic-field related studies performed up to now were free for all according to the target position, magnetic field strength, stimulation number, stimulation duration, and so on. Since these magnetic field stimulation systems were not verified, the quantification process of expression effects of the magnetic field. In this study, we developed a magnetic field generation system for magnetic field stimulations to make biophysical influences more quantitatively accessible. This system was designed to adjust the magnetic field strength and frequency to find out an optimal magnetic field stimulation conditions for chondrocyte proliferation and adipogenesis. Computer simulation was conducted to predict the uniformity of the magnetic stimulation strength over the cell specimen area, and it’s real intensity and uniformity were measured using a gauss meter. Also, the appropriateness of proposed magnetic stimulator device for the cellular level study was confirmed by observing the change of biological effects, that is, cell proliferations of chondrocytes and differentiation of adipocytes. Three types of coil combined with Helmholtz and solenoid coils were designed to generate a homogeneous magnetic field in a cell sample. And a magnetic field waveform control module including high current driver circuit was suggested to generate a time varying magnetic field of 100Hz or less, magnetic field output in the range of 0.3 T to 1.0 T. This system was used to evaluate the effects of various magnetic field stimulation on the proliferation of chondrocyte stem cells and the differentiation of adipocyte derived from hMSCs. The stimulation protocol effective for the proliferation of chondrocyte stem cell is 0.3 T/10 Hz/60 min, and time-varying magnetic stimulation protocol effective for to the differentiation of adipocytes drived from human mesenchymal stem cells is 0.3 T/10 Hz/10 minutes, which shows a significant differencecompared with the group without magnetic field stimulation. The stimulation protocol presented in this study showed effective results with different protocols for chondrocyte proliferation and the differentitation of hMSC. This suggests that other type of the magnetic field stimulation is necessary depending on the object. Therefore, it can be concluded that a device like the magnetic field stimulation system proposed in this research is needed to analyze human body and cell change by applying magnetic field stimulation
