Abstract To expand the consumption and industrial use of potato-based food ingredients, this study was conducted to design the non-thermal process to separate individual parenchyma cells from potato whole-tissues, to prepare the dehydrated potato parenchyma cells with ungelatinized starches using the developed isolation methods, to determine the main factors affecting the isolation yields of potato parenchyma cells with respect to hardness and chemical compositions of potato whole-tissues, to investigate the physicochemical properties of the dehydrated potato parenchyma cells, to develop low-glycemic food materials of the dehydrated potato (termed as potato granule) through chemically-crosslinking reaction with citric acid. Using whole-tissues of domestic potatoes, the individual parenchyma cells (PC) were enzymatically isolated. The frozen potato whole-tissues exhibited 2 times higher yield of individual PCs than raw (untreated) counterparts. Commercial pectinase mainly comprised of pectin lyase was 200 times diluted and added to the reaction medium. The thawed potatoes were combined with sodium citrate buffer (100 mM, pH 2.5) to 1:4.6 (w/v) mixing ratio. 400 ppm of ascorbic acid and diluted pectinase was added to the mixture, after which it was incubated at 50℃ for 2 h under the continued stirring at 100 rpm. The maximum yields of the individual potato were 66.1%, based on the dry matter of their thawed whole-tissues. Among its 24 domestic cultivars, Chuyoung, Chudong, Chuback, Gawon, and Dejima potatoes were appropriate to produce the dehydrated PCs. The dehydrated PCs (relative to potato flours) obtained under the optimal conditions exhibited lower crude protein and crude ash contents. Relative to potato starch and flour, the dehydrated PCs revealed higher swelling powers up to 55℃, but lower swelling powers over 55℃. Their gelatinization temperatures were higher relative to potato starches and lower relative to potato flours. The dramatic viscosity development of potato starches and flours were not observed for the dehydrated PCs. Considering overall results, the dehydrated PCs would be greater potential of resolving their shortcomings revealed in industrial applications. Without heat treatment in high-moisture food system, the developed dehydrated PCs showed lower digestibility than potato starches. However, subjection of dehydrated PCs to heat treatment exhibited higher digestibility. Accordingly, the dehydrated PCs was not likely appropriate for food ingredients to modulate the glycemic response. Nevertheless, in low-moisture food system, the digestibility of dehydrated PCs (relative to potato flours) be limited. Consequently, the dehydrated PCs still modulate the glycemic response for low-moisture foods. The non-digestible materials of the dehydrated PCs were prepared using citric acid cross-linking reaction. The citric acid-crosslinked potato materials revealed the lower rapidly-digestible starch (RDS) fraction and the higher resistant starch (RS). Accordingly, citric acid-crosslinked PCs may have the greater merits on its industrial utilization as a food material to control the glycemic response.
