Diverse fungal endophytes isolated from mountain cultivated ginseng (MCG, Panax ginseng Meyer), a traditional medicinal plant, were explored for their diversity and investigated their potentially biocontrol activity against ginseng pathogens Alternaria panax, Botrytis cinerea, Cylindrocarpon destructans, Pythium sp. and Rhizoctonia solani. A total of 1,300 fungal endophytes were isolated from disease-free and surface sterilised plant segments (root, stem and leaf) from MCGs grown in 24 different geographic sites. 129 fungal isolates were authenticated by molecular identification based on rDNA ITS sequence analysis. The fungal endophytes belonged to Ascomycota (81.7%), Basidiomycota (7.08%), Zygomycota (10%) and unknown (1.15%), and classified to 59 genera. Analysis of diversity indices across sampling sites suggested species abundance as a function of geographical and environmental factors of the locations. Assessment of 129 fungal isolates for their biocontrol traits to 6 ginseng pathogen showed that ethyl acetate extract of T. polysporum demonstrated the highest activity against all pathogen. In contrast, T. citrinoviride showed antagonistic activity with mycoparasitism by means coiling around hyphae and penetration against all ginseng pathogens. We found that T. citrinoviride produced highest activity of endo-1, 4-β-D-glucanase, and it caused down-regulation of B. cinerea genes related in growth and virulence (BCG1, BAC, BOT1, PKA1 and TRE1). Inoculation of ginseng plants with T. citrinoviride by dipping inducted ginseng PR genes (PR2, PR3, PR4, PR5, PR6 and PR10) and ginsenoside biosynthetic genes (FPS, SS, SE, DS and PPDS), and also hormone levels (ZA, ABA and JA). The ginsenosides (Rb2, Rc, Re and Rg1) also increased after inoculation of T. citrinoviride. By the real-time PCR quantification of T. citrinoviride, successful endophytism was confirmed. In planta assay, T. citrinoviride inoculation significantly reduced the disease caused by B. cinera and C. destructans in ginseng plants. With this biocontol potential, we developed formulation agents as DP and GR which have been considered to convenience and stability, and those agents were also showed biocontrol activity on B. cinera and C. destructans with ginseng plants. Inoculation of MCG plants with DP and GR by foliar application enhanced disease resistance and gisenoside concentration (Rb1, Rb2, Rc, Re and Rg1) in field tests. These results indicate that T. citrinoviride is a beneficial endophyte with biocontrol properties, and it can increase the quality and yield of MCG plant.
