Microcystis aeruginosa, a freshwater microalga, is capable of producing and accumulating different types of sugars in its biomass which make it a good feedstock for bioethanol production. Different factors increasing growth rate and carbohydrates productivity of M. aeruginosa was investigated. Also, efficient pretreatment methods for the biomass of the M. aeruginosa to convert to bioethanol with enhance yield was experimented. The whole cell extract was evaluated for antioxidant potential. Also, the synthesis of bioactive constituents including the toxic metabolites, microsystins were induced, extracted and investigated for antiangiogenic, antimicrobial and anti-obesity activities. The wild type M. aeruginosa was mutated with gamma radiation for increasing the carbohydrates utilities. MF media (modified BG11 media) and additional ingredients e.g. aminolevulinic acid (2 mM), lysine (2.28 mM), alanine (1 mM), and Naphthalene acetic acid (1 mM) as cytokine promoted M. aeruginosa growth and sugar contents. Salmonella showed growth-assisting effect on M. aeruginosa. Enhanced growth rate and carbohydrates contents were observed in M. aeruginosa culture grown at 25°C under red LED light of 90 μmol m-2s -1(5000 lux) intensity. Under the above conditions carbohydrates rich biomass of M.aeruginosa (O. D660 2.4) was obtained and was efficiently pretreated by the combination of lime, acid and enzymatic hydrolysis. A sugar rich algae juice (Glc16 mM/mL) was obtained and was used as a substrate in fermentation process. Highest yield of bioethanol (50 mM/mL) was obtained when Brettanomyces custersainus, Saccharomyces cerevisiae, Pichia stipites and Klebsiella oxytoca were used in combinations for fermentation process as compared to their individual fermentation. The M. aeruginosa whole cell extract showed antioxidants activities against DPPH, ABTS and superoxide radicals. The crude microcystins isolated from M. aeruginosa significantly inhibited the tubes formation of HUVECs in matrigel. RT-PCR analysis of the mRNA expression revealed down regulation of the angiogenic signal proteins VEGFR-2, PI3 K, β-Catenin, VE-Cadherin, Akt, and NF-kB in the cells treated with the crude toxin. The crude extract of the toxic metabolites exhibits anti-obesity activities by inhibiting the in vitro proliferation of 3T3 cells line. M. aeruginosa crude microcystins significantly inhibited the lipid accumulation in the 3T3-L1 cell line. RT-PCR analysis shown downregulation of the adipogenisis and lipogenesis regulating signals proteins (PPARγ, C/EBPα, and SREBP) by the crude microcystins. Furthermore, the crude toxin exhibits antimicrobial activities against seven serovars of the zoonosis salmonella and the fungus strains A. niger and F. solani. The blooming M. aeruginosa showed toxicity against nematodes. The overall results indicate the feasibility of M. aeruginosa as a feedstock for fermentative ethanol production and rich source of bioactive therapeutic agents for safe use as natural constituents in pharmaceuticals products.
