Exception for the bloom-forming period of Microcystis spp., Aphanizomenon spp. has been occupying a high proportion in phytoplankton population and frequently forming the bloom in the Nakdong River. The number of taxa in the Genus Aphanizomenon which is reported in the South Korea is extremely limited, and studies involving morphology but also ecology and physioecology on the taxa of this genus have not been conducted at all. Thus, in the Nakdong River, explaining the taxonomic criteria and the phenomenon of Aphanizomenon blooms based on previous studies is indeed difficult. The purpose of this study is to provide understanding the taxonomy and physioecological characteristics of Genus Aphanizomenon which is bloom-forming cyanobacterium in the Nakdong River. For this purpose, this study describes the morphological characteristics of the Aphanizomenon spp. and related species collected from the natural samples in the Nakdong River, investigates the morphological and phylogenetic characteristics, detects toxin-producing genes, toxin-producing ability, allelopathy effects and response of growth to varying temperatures and nutrient conditions on the strains of taxon which frequently cause bloom in the Nakdong River. The taxa from the natural samples collected in the Nakdong River were classified into 4 species under the genera Aphanizomenon Morren ex Bornet et Flahault 1888 and Cuspidothrix Rajaniemi et al. 2005 by light microscopy. The following taxa were identified: Aphanizomenon flos-aquae Ralfs ex Bornet et Flahault, Aphanizomenon klebahnii Elenkin ex Pechar, Aphanizomenon skujae Komárková-Legnerová et Cronberg, Cuspidothrix issatschenkoi (Usačev) Rajaniemi et al.. Cuspidothrix issatschenkoi are described for the first time in the Nakdong River. In addition, Aphanizomenon klebahnii and Aphanizomenon skujae are newely known to South Korea. The identity of toxin producers remains as only a hypothesis unless they were identified by strain isolation and analytical confirmation of both the cyanotoxin production and the genetic identity of the monoculture. 2 strains of genus Aphanizomenon which is increasing its frequency of appearance were isolated from the weir area of Nakdong River. Morphological features of the 2 strains (strain number: DGUC001, DGUC003) were observed (with naked eyes) as the shape of aggregated trichomes in parallel fascicles which can reach up to macroscopic size and a hyaline terminal cell without aerotope. In addition, the 16S rRNA phylogenetic analyses showed that the strains were identified as the same species with high genetic similarity of 98.4% and grouped within a monospecific and supported cluster I of Aphanizomenon flos-aquae selected from GenBank of the NCBI. The cyrA and cyrJ genes encoded for the cylindrospermopsin-biosynthesis were not detected in the present study. The sxtA gene was in amplified the two strains whereas the sxtI gene which had been suggested as a suitable molecular marker to detect saxitoxin-producing cyanobacteria was not found both strains. Thus, the two strains isolated from Nakdong River were identified as the same species of Aphanizomenon flos-aquae Ralfs ex Bornet et Flahault 1888, and the two strains were confirmed as potential non-producing strains of the saxitoxin and cylindrospermopsin. The two stains of Aphanizomenon flos-aquae were confirmed as non-toxin-producing strains because saxitoxin and cylindrospermopsin in intracellular and extracellular fraction were not detected by ELISA (Enzyme-Linked ImmunoSolbent Assay). The optimal growth temperature (Topt) of Aphanizomenon flos-aquae from the Nakdong River was 19.9℃ (18.3-21.2℃), and the temperature for growth ranged from -0.3℃ to 34.3℃. Specific growth rate (μ) in modified CB medium varied at 0.10-0.16day-1, and the maximum growth rate (μmax) was 0.17day-1. Although growth curves under N-existed and N-depleted condition were partially similar, growth under N-depleted condition indicated that there were relatively slowed specific growth rate (μ=0.09-0.14day-1) and decreased maximum cell density (385,826cells mL-1). However growth under P-depleted condition were restricted in all the temperature range. The algae inhibition tests were performed to assess the potential allelopathic effects of the strains' filtrates on the growth of 4 algae strains (Microcystis aeruginosa, Aulacoseria ambigua f. spiralis, Aphanizomenon flos-aquae, and Scenedesmus obliquus). The exudates or filtrates from the Aphanizomenon flos-aquae resulted in significant inhibition effects of algal growth on the Aulacoseira ambigua f. spiralis and Aphanizomenon flos-aquae (p<0.01).
