Indeed, early detection and definitive therapy are necessary to decrease the risk of metastatic disease. Somatic oncogenic mutations of v-Raf murine sarcoma viral oncogene homolog B glutamate for valine at codon 600 (BRAF V600E) have commonly been identified in primary human melanomas. Moreover, the development of an immunohistochemical stain for the BRAF V600E mutation has recently emerged as a potential rapid screening test for melanoma. Clinical interest in BRAF mutations has expanded the application of immunohistochemistry (IHC) for detection of BRAF V600E to a wide variety of tumor types. Therefore, in this study, we retrospectively analyzed the clinical records of 47 patients who had been treated for pathologically proven melanoma at Hallym University Sacred Heart Hospital from 2005–2015. The study compared the detection rate of BRAF expression through BRAF VE1 antibody and BRAF mutation in 47 paraffin melanoma specimens by two discrete methods of IHC and peptide nucleic acid (PNA)-mediated PCR. The correlation of BRAF expression with clinicopathological features of melanoma was also investigated, as was the sensitivity and specificity of IHC analysis using a BRAF VE1 antibody for detection of the BRAF V600E mutation in melanomas. BRAF mutation was not correlated with any clinicopathological factors of melanoma. Moreover, BRAF VE1 antibody IHC assay exhibited a 19.1% BRAF expression with high sensitivity (90%) and high specificity (92%). In conclusion, the study demonstrated that BRAF VE1 antibody IHC assay may be useful for detection of BRAF mutation in cutaneous melanomas. Accordingly, BRAF VE1 antibody IHC assay may be a rapid, cost-effective alternative method to conventional BRAF mutation testing that could allow BRAF V600E mutant cutaneous melanoma patients to be rapidly triaged into appropriate treatment pathways.
