또한, 실험관내 시험을 통해 인간 정상세포주인 제대정맥 내피세포(HUVEC)과 혈관대동맥 평활근세포(HaSMC)에서 10mM 농도까지 처리 했을 때 독성이 없음을 확인하였고, 혈당상승 억제 기전이외의 효능을 확인하기 위한 연구로 cell migration assay와 면역세포 활성 실험을 한 결과 세포의 이동성에는 영향을 미치지 않았지만 면역활성 실험에서 mouse의 비장세포에서 추출한 면역세포들 중 B 림프구 특이적 표면항원인 CD19의 발현을 대조군대비 10배 정도 증가시켜 B 림프구와 관련된 면역활성에 유용하게 쓰일 수 있다고 판단되었다.

The present study aimed to determine the large scale production and effects of arginyl-fructose (AF) supplementation on glucose control in the preclinical studies with animal models and clinical studies with prediabetes. AF could be effectively and simply produced only by the addition of 10% acetic acid at 80℃ with mixing in the 1:1 ratio of arginine and glucose. Inhibitory effects of AF on the proliferation and migration of human normal cells(HUVEC and HaSMC) were not revealed at concentration of 10mM. Interestingly, we founded that the CD19+ B cell population in splenocytes (of mice) treated with 10 mM AF was increased in a remarkable level (10 fold), compared to the control. This datum suggested that AF may increase the B cell-specific immune response. In addition, we have studied the inhibitory effect of AF on blood glucose level in diabetic db/db mice, a genetic model for type 2 diabetes. The db/db mice were randomly assigned to high-carbohydrate diets (66.1% corn starch) with and without AF (4% in the diet) for 6 weeks. Changes in body weight, blood glucose level, and food intake were measured daily for 42 days. Dietary supplementation of AF resulted in a significant decrease of blood glucose level (p < 0.001) and body weight (p < 0.001). The level of HbA1c, a better indicator of plasma glucose concentration over prolonged periods of time, was also significantly decreased for 6-week period (p < 0.001). Dietary treatment of acarbose (0.04% in diet), a positive control, also significantly alleviated the level of blood glucose, HbA1c, and body weight. These results indicate that AF Maillard reaction product improves postprandial hyperglycemia by suppressing glucose absorption as well as decreasing HbA1c level. Moreover, we performed preclinical and clinical studies to evaluate the toxicity of AF in in vivo. In preclinical studies, AF did not show any severe side effects on 13-week repeated administration. NOAEL (No-Observed-Adverse-Effect-Level) of AF was determined to be 5,000 mg/kg/day in both sexes. In clinical studies with the collaborative research, AF supplementation (1500 mg/day) showed a beneficial effects on control of glucose levels in the blood of human prediabetes group. Taken above results together, we expect that AF would be a potential anti-hyperglycemic material in the field of heath food research.
