DH-1 에 대해 다양한 유전자 조작 기술을 최적화하였다. 최적화된 유전자 조작 기술을 사용하여 phytoene desaturase (CrtI) 를 결손시켰다. 그 결과 세포의 색이 사라짐을 확인하였고 이를 통해 carotenoid 의 합성 경로를 규명하였다.

Methane can be utilized and converted through chemical processes, but they demand high costs due to the high temperature and pressure. To solve this problem, biochemical process using methanotrophs has attracted much attention. Methanotrophs can utilize methane as a sole carbon source and produced value-added products such as methanol, fatty acids, etc. However, the productivity of biological conversion of methane using wild type methanotrophs are very low. Therefore, genetic manipulation of methanotrophs are needed to increase the productivity of bioconversion. In this study, we developed genetic manipulation systems and using this methods, genetic manipulation of methanotrophs was carried out. First, various conditions of transformation techniques (conjugation and electroporation) were optimized for Methylomonas sp. DH-1. Based on this techniques, phytoene desaturase (CrtI) gene of Methylomonas sp. DH-1 chromosome was removed using double cross over system. As a result, the color of the cells disappeared and the pathway of carotenoid synthesis was identified.
